尼龙菌的移码突变T是由其自身一个质粒插入的,所以必须证明这个插入不是该机体生
命机能的一个intrinsic function,而是一个意外。
因为生命机能的内在功能里有修改自己遗传密码的部分以达到适应外界变化的例子,比
如人体的免疫系统B细胞,能辨认理论上的几乎任何蛋白质,其辨认过程就是靠自动自
发的DNA mutation,其mutation有固定的process,类似一种分子算法。辨认抗原编码新
抗体的过程就是机体的分子计算过程。
可以看一下下面链接的V(D)J recombination,和Somatic hypermutation and
affinity maturation两个部分。对B细胞,后者就是其B细胞自身在遗传(分裂中)改
变自身DNA某个设定区域的编码,这是其特定功能的部分。

其实验证这个比较容易,只要把尼龙菌放入前身细菌的常见生活环境看其是否可能还原

粒体DNA(mtDNA)必须是从女性到女性的。为什么?因为精子卵子结合的时候,只有精
子的染色体进入了即将形成的受精卵,所以只有不断地从女性到女性,才能继承这同一
的mtDNA(还要排除宇宙射线等不可避免因素造成的体细胞突变somatic mutation)。
所以“线粒体夏娃”和“Y染色体亚当”只能是假说而已。
纯粹地试图用科学来解释圣经,这个本身也是问题。为什么?因为科学本身就不能解释
很多的东西,比如政客们争论的全球气候到底是不是暖和一点了,再比如是什麽神经通
路控制人会在开Uhaul的时候拐弯半径会更大一点。所以用有限的知识/认识来解释无限
的外在内在空间,逻辑上是不合理的、是有系统误差的。
用科学解释圣经,更加地危险。为什么?上帝已经把自然科学和社会科学等等启示在圣
经里;同时,圣经已经教导我们如何理解圣经(包括自然科学和社会科学)。本末倒置
地试图利用有限的科学来“解释”圣经,逻辑上是不合理的。而且也是很危险的。为什
么?因为我们会问:我应该不应该相信现有科学无法解释的圣经部分呢?

Now, it's suggested to use somatic stem cells concept than adult stem cells.
As far as I know, progenitor cells are of less differentiation
potential(multipotency, just many progenies but not all), while stem cells are
of pluripotency (All of the progenies). So in this matter, PC is considered
more committed, determined, or differentiated than SC. However, most time ppl
find adult stem cells which are really progenitor cells.

个人觉得 KO 老鼠最能 mimic 生理状态了, 比如某些 tumor suppressor gene
的 condition KO 模型, 就是最好的mimic somatic mutation 导致肿瘤这个生理
过程. in vitro 的实验能解释一些事情自然是好的研究, 但是in vitro 的结果
不能在 in vivo 里得到证实, 或者结果相反, 那自然是垃圾研究.

还有 从ES细胞到终末分化的细胞,并不单单是转录因子的作用,
有时候细胞间的相互影响作用也很大

Theoretically, directional differentiation is easier than reprogramming,
because the latter is not a natuarl process. There is no any reason that
transcription factors should not force cells to differentiate into specific
lineage, given the facts of iPS.
I don't think any lab has achieve this, though.

在yamanaka之前,有这样的观念,用1-2个cDNA transformation,然后得到什么什么细胞系
这个本质是stable cell line和transgenic 路数
iPS 开辟的是reprogramming这个路线,在iPS之后,马上很多人就做了
transdifferentiation,依据同样的思路。
在Yamanaka之前,有些人搞过这个XXX细胞到iP,比如Stanford 胡继繁就有类似的想法,还申请了
专利,但毕竟没有成功。
当然,iPS有很多要细化的研究,比如gene deletion duplication trouble in the final iPS
iPS generated from somatic cell? or from few tissue stem cells in the
primary culture???
无论如何,这个是reprogramming的事情,是的,某些伙计说的,或许弄出了生物学有
史以来最大的reprogramming 泡沫,呵呵

问过一个教授,他说,要做人的话,必须ips和es来自于同一个人,这样才能排除
genomic DNA的variation,但这是不可能的,人取了es就不可能有ips了。
所以他说只能做mouse的stem cell。找同一strain的mouse,去es,再用somatic 诱导
出ips,这样比较他们的不同才有意义。
大家说说看,这种说法对吗? |
