The protein samples was digested by trypsin, which cuts after K and R. But
why some peptide fragments follow amino acids other than K and R in the
result file? and the same fragment appears more then once. are those
fragments real?

binding profile. By comparing control and mutant groups, I am hoping to
find targets that are more disease relevant. I use magnetic beads for Co-IP.
Now here comes the questions:
Would it be good enough to just use SDS buffer for elution? I know this is
the case if my next step is western blotting.
If not, would an acidic glycine buffer followed by a basic buffer (Tris-HCl

proteolytic cleaved 的片断。倒是SDS_PAGE其实是很不准确的，因为跑出来的结果和
蛋白的形状和修饰状态都有关。

还有，通过sds－page来判断蛋白质大小的这种方法太不准确了，有些蛋白（没有任何
posttranslational修饰）在page胶甚至可以表现出2倍于理论分子量的size，所以你不
能说这个33kd的蛋白质就一定不是你的target protein。
当然size上的不一致也有可能是蛋白的修饰引起的，尤其是在细胞里提取的蛋白

phd的时候没有听说过这个career path,毕业以后在工业界工作过一段时间,拿到绿卡就
马上开始申请tech spec的职位的.主要的考虑是早点拿到绿卡以后再转行比较踏实.
因为我知道有人phd毕业就直接找这样的工作,所以我在想,这个"等绿卡"算是合理的"
phd毕业没有马上进入IP law领域"的理由吗?

且如果他们已经有外国人雇员，他们很有可能知道在律所申请绿卡是多么费事。
我是用的这个理由，没有觉得好像有太大问题，以后也会接着用，事实上也的确是这样。
顺便搭车问另外一个面试问题。如果被问到什么时候能开始工作，怎么回答比较好。
还在做实验，基本差不多了但是paper还没有开始写。老实说我是不太在意了，所以有
一次我回答说我会给工作priority,基本上可以任何时候开始。这样说会不会让人觉得
不负责任？还是说给我几个星期把工作做完比较好？我猜一般tech spec program招人
不是说想尽快找个人来干活的吧？ |

给做，是办EB-2还是EB-3，EB-2现在看起来快一点，但是EB-3还是很慢，NIW值得试一下
身份的问题面试我建议是要问的，如果你需要H1B应该提出来，不然你也不能去工作呀
，绿卡每个所可能都不一样，我觉得跟你是不是上law school没啥关系，你如果能进这
个所，H1-B办完以后半年到一年的就可以跟老板谈绿卡的问题，如果他们想留你会考虑
你的要求的，看起来你面的这个所本身就有移民部，如果所内就能把你的绿卡办了就更
方便了，关键是你能拿到offer，绿卡都可以进去以后再商量
