truth, i think so ... well, don't tell boss and buy another
vial then. next time, try not to do that. i suggest you use
a cell storage from Nalgene. add isopropanol into it, put in
the vial with cells (cells are in DMSO and FBS), and put it
@ -80C. it is said to decrease the temp by 1C/hr. after
put it in the -80C two days, you can now transfer it to
liquid N2. when you thaw the tube from N2, put it in 37C
water directly, don't stand outs |

(i.e. Buffer) and 27.5G needle (i.e. Needle)
1. wash cell with PBS, spin down cell, remove all PBS.
2. resuspend cell in Buffer, pipete 10 times, sit on ice for 15 min.
3. use shringe and Needle, pass the suspended cell through Needle 7 times
4. site on ice for another 10 min.
5. pass through Needle another 7 times
6. spin at 500 g to remove debris, 100k g for membrane fraction |

the protein property. If you know it is an enzyme, I think you had better add
glycerol, because freezing and thawing cycle can kill the enzyme. For other
proteins, I do not bother to add glycerol either.
And to get rid of gluathione, besides dialysis, there is an easy way. Take a
spin-column with an appropriate cut-off filter, e.g. 30KD, spin, concentrate
the protein and get rid of the solvent in the meanwhile. Then add |

FBS里面有一种叫做fibrin的蛋白质，thaw之后，就会聚集，产生蛋白质颗粒
（black dots），随着培养时间的延长，就会聚集越多。不同批次的FBS，
不同品质的FBS，里面含fibrin的量略有不同。
不要把这些蛋白质颗粒filter掉，细胞需要这些蛋白质，因为可能有生长因子。
实际上，细胞很喜欢吃这些蛋白质，你会发现很多细胞里有蛋白质颗粒。
如果颗粒实在太多太大了，可以非常低速地离心一下，去除掉过大的颗粒就行了。 |

protein不会被degraded，至于posttranslational modifications，test过的种类不多

phosphorylation of a protein, why not use western? Even if you do ELISA to
see p-protein, no need to sonicate....RIPA is strong enough to get all your
cytosolic and nuclear protein out. Sonication mostly is used to break down
chromatin, DNA or chop down plasma membrane to release cytoplasma membrane
protein....If I am wrong, please someone corrects me.. So I think in your
case, you probably don't need sonication at all.
