My boss want me to find out exactly how many protein kinases (among them,
how many tyrosine kinases and serine/threonine kinases) and lipid kinases in
human beings.
I chekced several references and a website " www.kinase.com'
I got the follows:
1, 518 kinases. 90 are Tyrosine kinases.
I cannot find the number of serine/threonine kinases and lipid kinases, even
though i searched many published papers.
Roughly, the protein kinase was mainly classified as TK and STK, but there

D. 100% sure. the key word here is "cytoplasmic enzyme". some protein
kinases require phosphorylation at serine/threonine for activation.
A is for membrane-bound protein.
B &C for extracellular (secretory) protein.
E for example mannose-6 P but not in Serine or Threonine .

(2)既然说到了氨基酸,那就再仔细看看氨基酸。
人体有8种氨基酸是不能自己合成的,必须通过外界摄取,所以叫essential amino
acid
其中这三种,目前食谱是肯定缺乏:Leucine,Threonine,Phenylalanine
分别有关:肌肉合成;免疫抗体合成;肾上腺素前体合成
还有有一种可能缺乏:Valine--保证愈伤,和氮代谢
而这几种重要的氨基酸,都可以从一种食物中获取:花生。
这其实就是问题核心所在。很多靠吃素减肥的人,很害怕加坚果。觉得“油”是很增肥
的东西。
所以猛控油,猛控油。你也explicitly的说了,你要“低油少盐”
炒菜油是应该低。但是天然食物中的油,不能一点没有。否则氨基酸和不饱和脂肪酸都
没有来源
给血液指标带来很大的危险。这个问题也和第一个问题correlate。
三大能量物:脂肪,蛋白质,和糖,必须得至少吃一个。所以我个人不同意把前两个都
给压到最低
一旦这样做,糖必然超标,只要你吃700卡以上就超 (目前食谱约1100-12
00)

如果你表达的是kinase,很容易自己修饰自己。
如果你表达的非kinase, 有少许可能被修饰,可能在Serine, Threonine, Tyrosine,
Histidine, Aspartate被修饰。
一旦被修饰而你又很介意的话,可以co express一些phosphatase, 或者invitro
dephosphorylation.

看看是不是SNP.
表以为别的species是S, 你这儿就没有SNP。
S变成T的SNP是常见的。
现在Genome测得多了,所谓的wildtype是相对的,没有绝对的wildtype.
所谓的reference 其实是most common variant,不是绝对的wild type.

又测序了一个克隆,目前为止没有突变.看来还是polymerase的问题.
问题是这个基因有2.5kb,还得接着测,但愿没突变了.

把突变那个和没突变的做个比较就知道要不要紧了啊。
没突变那个是不能省的,要不就说不清。
多设计几个引物,多挑几个克隆,一次全测了,不要挤牙膏啊。

大家对我一次测一个克隆这么有意见啊,呵呵,我等测序的时候还干别的有很多事啊,不
适只做这一个实验呀.不过已经拿到了测序结果了,克隆没问题可以接着往下做了:)
其实同时还做了一个5k的,主要是测这个耽误了两天.

同学,现在实验时间往往比钱更重要,如果你做的是热门课题,至少有3个顶级实验室
同时在做这个,你只要晚一点就被scoop。
我们做实验验证的时候,同一个PCR基本用3种酶同时扩增,有一次(第一次测序失败)
同时用了3种酶,两侧各3个引物,27个PCR,54个测序一起做。
