h*******n 发帖数: 2052 | 1 I plan to perform in vivo immuno-receptor blocking with antibody and examine the effect of host-defense.
Where can I find the information below:
1. Ways of injection (i.v., i.p., etc.)
2. Dose of each injection
3. How often do I inject the mAb?
4. For how long do I inject the mAb?
I cannot find any paper describing similar experiment with my specific mAb.
Any suggestion is appreciated. | c*****i 发帖数: 1392 | 2 all your questions are dependent on which receptor you are trying to
block....
examine the effect of host-defense.

examine the effect of host-defense.

depends on specific cell types your receptor is expressed on
you need much less if you are targeting circulating cell by i.v
depends on the first question.
if i.v., I would say 60-80 ug would be a good dose to start with.
It also depends on the antibody though.

depends on the nature of your experiment
Chronic or acute

1. iv is more for acute reponse. ip is slow but maybe last longer. it
depends on what receptor you are looking at and what tissue are they located
2. somebody probably has done that, or the data sheet of your mAb suggest a
starting dose to try. you might need to do a escalating dose to find out the
optimal dose
3. again it depends on your specific receptor, their turnover rate, the
halflife of your mAb, etc...
4. often peopl
