If you want to use some mammalian cell line (such as Hela) to produce and
purify protein to mg quantity,
(assume you can not get it from E coli, yeast, insect cells, or the protein
has some post-translation
modification that only occurs in mammalian cells) you'd better have the
protein secreted into the medium, so
that 1) yield will be higher; 2) less toxic to the cells.
If you just want to see how the cells behave/react to the high level
expression of the protein within the cell, it
would be easier.
