厚？我本想切100nm,可是现有的microtome在这个设定下很不稳定,一会厚一会薄的，那
个technician告诉我要切
200nm才行，可是那样就太厚了，电镜下看就是一团黑。
新手一个，刚开始摸索，请教各位给点microtoming方面的建议，特别是对polymer
membrane类的样品。我是把膜
mount在epoxy glue里切的，用的是glass knife，不是diamond knife。 |

这个是不是材料deform造成的. 冻得不够结实?
以前也用epoxy切过膜,感觉如果膜很薄很软,切的时候经常被从epoxy里扯出来,十分不

coating on the sample section on the Cu mash.
diamond is better than glass, however before you try diamond you can first
try water+glass if your film is stable in water
cryo cut you need to use half water + half dmso
you need patience + luck

发帖数: 38 | 8 谢谢你的建议，那个polymer membrane在TEM，200KV底下经常会移动，是不是就是不稳
定？我只知道用SEM时，
不导电的sample要先coat一层Au或是Pd，要carbon coating的话是不是将有sample的
copper grid 放到相似的仪器
里再涂一层carbon呢？

把epoxy给烧了变成黑黑的
一团团的东西附在你的样品膜上面？
我经常看到这些小黑团，开始还以为是我样品里的东西，但是后来发现不太象，所以怀
疑是那个epoxy glue，但是还
没有确定，就想问问有没有发现类似现象的？。。。

staining.
there are two types of contrast to distinguish ur sample, check the mass or
thickness contrast |

。能再具体一点说说mass

发帖数: 28 | 13 1. 样品能溶于什么溶剂不？试试SPIN COATING， 然后把膜转到TEM GRID上面；或者直
接ＤＲＯＰ ＣＡＳＴＩＮＧ在ＧＲＩＤ上面；
2. 200nm 的膜用 200KV的TEM 看应该没问题， 我这儿一个很老的TEM升到120KV的时候
可以看500 nm的膜； 是不是机器不行了或者你没调好STIGMATION
3. 玻璃刀也可以切１００nm以下的膜， 不过要多试几次， 在刀口坏掉之前应该能
搞到一两片好膜 |
