I once changed one Gateway laptop CPU. Bid on ebay and got one
pentium-m 1.3G for $8 (shipping included).
Then sold the celeron-m 1.5G for $30 ("buy it now...")
The speed is about the same. Pentium-m is a little faster.
Battery life increased from 3hr to 4hr. :-) |

我的电脑本来是Windows 8.0（本来就很讨厌这个版本的界面了，不会用）， 最近老自
己跳出来升级成8.1的要求，我不想麻烦，就一直选‘Later“ 往后推，有一天它也不
问，自己就开始重启安装升级了，可是升了一百年还只完成80%就再也不动了，我把它
硬关了，再打开，它又重新开始，还是到80%就不动了，最后它自己说：can not
complete, will restore your previous version.然后就出现底下一根空白蓝条（就
是本来有控制面板，音量，输入法哪些东西的那个位置），上面的屏幕什么也没有
。我吓死了，还以为电脑坏了，又硬关，再打开，一切又恢复正常了，还是8.0。
我还挺高兴，结果今天又问啦：你想什么时候升级呀（选：1hr,2hr,3hr,4hr)？我的天
，难道上次那堆乱七八糟的又要重来一趟，耽搁好多时间不说，我觉得电脑都快坏啦。
为什么就没有一个选项是：我不想升级！而且这次连”Later“的选择也没啦，唉。。。
求大虾们不嫌问题太弱，指点指点吧！太感谢了！ |

不过那天就把三章草草的看了一下。
平均每章三个小时左右吧。
第二天考试是下午1点。我就早上起来把课后习题看了一下。
两个半小时吧。
照你说的情况，应该是AUD最难。
因为做过审计的话那些substantive testing和SOX的东西就好理解得多，否
则的话有些概念还是比较抽象的。
我觉得复习AUD主要理解，大部分内容还是讲逻辑的。
如果理解了什么科目为什么要那么审计，那就不需要费那么多脑子去生记了。
所以话说回来，还是根据个人情况。
做过审计的肯定不会觉得审计最难，
而平时做税务工作的肯定觉得REG简单。
对于在校学生的话我觉得大概AUD最难。
至于复习时间，我是这么分配的。
看书 - 普通章节每章3小时左右，陌生章节每章4-5小时。
这样大概占据了80%-85%的复习时间。
看完以后再花一点时间做课后题以及看答案。我做题的时间大概是：
BEC - 3hrs，AUD - 5hrs，REG - 10hrs，FAR - 5hrs
（只做自己缺少把握的章节，每章半个小时到一个小时）。
因为时间紧，我Homework Reading基本都没看（四门加起来看过两 |

since you have no intermediate & advanced accounting background
2) Spend some money, FARE changed a lot since 2007, get the latest review
material, (BECKER is a good review material, $600 for FARE)
3) If you decide to use BECKER, study the review lecture first, read the
text books for at least 2 times.
4) Practice the after class questions for at least 3 times.

since i don't know the induction condition you use, here is my induction
condition for the protein I tried.(23 du, 2-3hrs) if still not good, try
18 du.
The proteolysis is inherent in GST fusion protein preps. You always get a 30
Kda form which is GST. The other problem about GST is that it is easy to form
dimer, especially the protein you work on is also easy to form dimer.

具体说说我的virus载体和transduction的过程吧~
1> retrovirus用的是MSCV based带有mir155 backbone的一个载体（某postdoc自己构
建的）有GFP marker。因为它只侵染dividing cell, 所以就在collect BM的当天做一
次spin infection （用fresh virus containing sup from transfected 293T cells)
，之后两天各做一次spin infection，然后让细胞在有GMCSF的medium里面继续分化到
day7 (中间换一次medium). 最后收细胞染色FACS. 现在结果就是no virus control分
化没什么问题，通常我得到90％ CD11c+细胞，其中MHCII+大概15％；问题是无论是
control virus (empty vector)还是有target shRNA的virus，细胞就不表达CD11c了 (
<10%），MHCII到没怎么变，还看过CD86之类的co-stimulatory molecul... 阅读全帖 |

我做了两次，基本是一致的结果，目前还在做。
用的是sigma的M2 flag agarose beads,
600-1000ug总蛋白做IP，pre-clearance with IgG beads for 3hrs,
add flag beads, 4度rotate for 5hrs,
wash 3 times with lysis buffer,
then wash once with high salt(500mM NaCl) lysis buffer,
final wash with TBS,
SDS sample buffer elute(下次准备用flag peptide elute)
还请多指教。

-14个钟头。有点拖拉，08年底决定考试，当时还在postdoc，期间经历发文章、办绿卡
、怀孕生子，总算一切搞定。先生在湾区找到新工作，就辞职搬家，全职复习。感觉可
以全职复习真是太幸福了，否则我好像总是差那么一点点没有ready。非常佩服那些一
面工作一面考试的战友们，因为我知道那有多么挣扎、多么不易。

-Took Kaplan 5 days course, very helpful! It helped me identify my weakness
and strengthens. My stress level was way down when I found out that I only
failed ICE part on the MOCK exam. I did not prepare at all when I went to
take the course.
-Read FA and core cases for less than 3 weeks. so total less than 4 wks
