现在不吃零食了, 吃的那个flaxseed oil 和 biotin 里面都有gelatin and glycerin, 不太清楚会不会有问题诶. 这两种是过敏了以后好几周才吃的, 因为医生建议吃点fish oil/flaxseed oil 的补充omega 3.
现在除了处方粮, 药和这两种supplements, 其他啥都禁了. 对了, 还吃一点blueberry, 也是医生说可以的... |

发帖数: 4970 | 13 如果红的太厉害要破了， 我就涂有cortisone 的cream /spray. 只要不舔下去就没有
什么副作用， 就是没有cortisone 口服效果好。 shampoo 是virbac 牌子的， amazon
上有， review 不错。
我家的也不知道是什么过敏， 所以我又洗澡又换粮又喷药的。 不过医生说
environmental allergies 的可能比较大， 因为一吃激素就有立竿见影的效果。 如果
是食物过敏一般没那么快见效。
现在就在一些比较好的牌子里轮流grain free, bison/venison/salmon 的狗粮， 加
costco 买的salmon oil， 吃激素的时候医生建议给的biotin也有时候吃点。不过我经
常给些有鸡肉什么的treats 好像没啥大反应。 时不时会有一大片hotspot 出来，喷药
水就行了。
现在很悲剧的我家猫咪也过敏， 脖子上一片包， 奇怪的是我们出门玩， 没人没狗，
她就自己好了。 一回家又开始长包了。。。。 |

首先是衣服:首选白大褂,老师一问，你就说这是你的家常衣服
例如:老师,我除了这个就没有别的外套了
老师://感叹?多么敬业啊
其实是打扮:越土约好
男生:头发像鸟窝(最好里面还放几个鸟蛋)
上衣:至少要三个月没洗
裤子:至少两个月
袜子:最好基本上没有洗过
鞋子:买来以后就没有洗过的,最好是天天穿的
女生:短发参看男生,长发不要梳理
最忌讳用化妆品,千万不要掩饰自己的豆豆
至于其他,偶不是很有经验,可以去问***

targetting DNA. Generally I believe the concentration of your probe should be
enough for the detecting because 10ng/ml of a 16mer oligo aproximately equals
to 1.5nM by molar concentration, which could capture 1.5pmol targetting DNA if
it bound at a ratio of 1 to 1.
To label or not to label, it depends on the method you used to detect the
targetting. If you use, for example, biotin to lable to probe, then label the
probe |

making non-radioactive probe incorporate only a small unit, like a biotin,Dig
or fluorescein into the probe via UTP or others instead of directly incorporate
the enzymes for detection,like HRP, AP. Since it is not easy to keep the full
enzymetic activity during the long experiment procedure.
So if you use klenow to label the probe, you can deactivate the klenow by
heating the reaction at 75C for 20min in presence of 10mM E |

Actually, if you can bio label or p32 label by yourself, there is no need to
buy such expensive kit.
For biotin labeled, 1 percent SDS can be used for blocking and washing. |

提纯，结晶都没有这些问题。现在是用SDS Sample Buffer 洗Biotin Beads并浓缩后有
的这问题。
此外什么样的蛋白样品容易有pH的问题? |

我就是看说明书上这么说的：
2.2 Release of Immobilized Biotinylated Molecules
The biotin-streptavidin bond is broken by harsh conditions. 5 mins
incubation at 65°C or 2 mins at 90°C in 10 mM EDTA pH 8.2 with 95%
formamide will typically dissociate >96% of immobilized biotinylated DNA.
Alternatively, boil the sample for 5 mins in 0.1% SDS for
protein dissociation.
Please note that proteins will be denatured by such treatment and Dynabeads
Streptavidin can not be re-used.

dynabeads，发现conjugation efficiency远低于预期值。而且比较头疼的是
要测出真实efficiency也很tricky。间接测反应后溶液里剩多少protein很
不准。
请高手指教有没有efficiency比较高的beads？最好是magnetic的（是不是
只有dynabeads系列？？），其他的介质也可以。最好是covalent linkage，
biotin-avidin的也行。BTW我的target protein pI在5~6

行，但是他们的人没有太关注rna。还有个macs什么的公司，有专门的kit。不知道效果

关于protein-beads ratio，我labmate用的方法是：先用少量beads，
bind protein at highest coverage，这个batch用来做beads可能带来background
的实验。然后recycle supernatant，加过量beads，把能bind的protein都bind上。
感觉比较make sense，就是有点麻烦呵呵。
btw，以前用M-270 carboxylic beads，现在改用这个，发现capacity低很多，我
随便就要用几百个microliter。。。

Streptavidin是链霉菌里提取的，是弱酸性的。
Neutravidin是化学修饰过的Avidin，是接近中性的。
Streptavidin和Neutravidin的优点就是non-specific binding比Avidin少。
不过你既然用的就是Streptavidin，那我就不清楚了。但是你的probe还是有可能跟
Streptavidin有cross-reactivity的。你screen probe之前用Streptavidin做过
negative selection了么？ |
