handle the cells carefully enough, you don't really have to coat the plates.
When you changing medium, do one plate per time, and don't try to change
medium for more than 3 plates in one shot. Cells don't like being exposed to
air for too long. Another thing is to, add medium or PBS to the wall but
not directly to the cells. |

处理时间过久了会有影响吗，比如在incubator里面放过夜了？

attachment culture in suspension?有没有人试过用irradiated MEF，beta cell会不
会自己形成一个小colony像IPS那样？实验室就我一人做diabetes的课题，没有太多经

瘦的，很多小泡泡。PCR检测是支原体污染了 。。。。
扔掉了50个15cm的dish的ES细胞 。。。。
浪费了好多试剂和时间啊。
请教到底该怎么避免支原体污染？是不是我操作的时候不小心？要注意哪些事情？
我看实验室还有人不带手套伸进超净台传细胞呢 。。。
我还需要扔掉哪些东西啊？培养基，trypsin，gelatin，都需要扔掉吗？ |

基本上都掉光了。因为切片是学校实验中心的一个人切的，她告诉我用的是Haupt's
gelatin adhesive。她做的片子给我基本上都掉得厉害，但有时候放在37°烘箱内多烘
一夜会稍微好一点。我现在买了poly lysine coated玻片回来，但不知道还有什么应该
注意的地方：比如说展片用水还是甲醛？烘片的温度，时间等。
另外，有人说做之前用60°烘片一个小时，会不会把蜡都溶了，rna搞降解掉？
请各位不吝赐教。 |

发帖数: 23 | 20 现在养的细胞在玻璃的coverslip上长的形态不好，可是在culture dish里没问题。在
玻璃coverslip上附着了poly-lysine/collagen/gelatin也没有太大帮助。想试一下
plastic或其他材质的coverslip/slide, 但查了一下好像一般autofluorescence比较强
。由于后续要做荧光染色，需要用到FTIC/rhodamine/DAPI，普通的plastic可能不行。
lab-tek有一种permanox plastic chamberslide号称minimal autofluorescence，不知
有人用过没？如果有人知道其他材质的plastic coverslip/slide适合用于荧光染色也
麻烦推荐一下。非常感谢！ |

被大肠杆菌污染了。我提了Total RNA，然后比较了，没有做分化以前和污染后的oct4
的表达水平，发现污染后的样品中oct4的mRNA水平比分化前有将近3倍的increase。
各位大牛，如果这个结果是对的话，是不是E coli也可以做reprogramming。 |

重 ,“这个是‘海澡酸纳’, 份量不能错, 所以要关风扇进行 ”。然后倒入一
盆温水内, 用玻璃棒不断搅拌, 约一个半小时后一盆晶莹剔透的蛋白物体呈现眼
前。她继而用少量化学物品“明胶”用水拌匀, 然后添进蛋白再搅, “明胶可以
令蛋白的浓度适中, 但因长时间搅拌会产生好多气泡, 要等十个小时才能散得完,
今天就先教到这里了”
第二天早上, 女导师再把适量的“酸纳脂”、“苯甲酸纳”、“邻甲基织维
素”及“白矾”等其它化学品一起混入, 但没有解释其作用, 三小时后“蛋白”
部份正式完成。
接着女导师开始教造蛋黄, “蛋黄其实就是蛋白, 只是加了色素的原因。”
她将部份蛋白状物体倒在容器内, 加上“柠檬黄色素”, 搅了几下就变成了鲜黄
色。
至于假蛋如何化作蛋形, 其中奥妙, 原来全是靠一个塑料模具, 以及含有
“氯化钙”的“神仙水”。塑料模具有两个凹位, 一个椭圆形, 一个圆形。女导
师边做边说：“圆形的造蛋黄, 将蛋黄倒入模具, 再倒入混有氯化钙的水在面上,
就会产生一层薄膜, 包围蛋黄就可以定型, 留意看, 现在变 |
