1，这两种细胞不会mix成一个pool，在组织学上有distinct的location
1.1 已知其中一种细胞表达的exclusive的marker，这种marker又有Cre Knock-in 小鼠
avaialbe, 那么可以用genetic lineage tracing的方法，通过观察Cre-loxp reporter
表达，看reporter是否会在另一个组织里出现。（利用Cre-loxp irreversibly mark
cell的特性）
1.1 不知道exclusive的marker， 可以用retrospective clonal analysis, 具体见这
篇 http://circres.ahajournals.org/content/early/2012/08/01/CIRCRESAHA.112.271064.abstract 主要思路是随机标记，
看你所说的这两个组织是否会被同时标记。
2,这两种细胞mix成一个pool

lineage分化的成体干细胞的分化出的一个lineage里表达另外一个lineage specifier
细胞最可能的选择是死掉，要是不死的话就处于一种lineage confusion的状态，但这
种状态必须在epigenome不稳定的情况下经过selection才能发生。如果两个lineage
specifier在一起时细胞更容易造成epigenome不稳定，并回复到原始的pluripotent状

driven的标记方法和实际mRNA/蛋白表达水平的差异也是个问题。虽然目前lineage
tracing被广泛运用，但我个人是非常反对把lineage tracing做为“金标准”的。
关于干细胞的研究，比lineage tracing更说明问题的应该是transplant之后系统的重
建。例如要证明细胞是ESC，最关键的标准不是Nanog等几个marker的表达，而应该看是
否细胞注入blastocyst之后能发育成chimeric mouse； 同理，证明hematopoietic
stem cell的关键在于是否能通过serial transplantation 来repopulate blood
system.类似的目前在gut,lung和skini等里边干细胞也都移植重建成功了。这在理论上

lineage tracing是看的homeostasis, transplantation看的更类似于injury repair.

的方式逐级重建整个H-lineage 。也由此self-renewal,multipotential 成为干细胞的
定义。
但实际上别的organ的干细胞并非如此。self-renewal是当然的。可multipotential 这
可真未必了。unipotential的就不能叫stem cell? 我认为这个定义需要修改。实际上
在现在很多stem cell研究里multipotential已不再是评价指标之一了，尤其在lineage

看. 一般来说Oct4啥的4天以后就降的很快了，然后lineage marker才慢慢出来。但是
因为是一个mixed population，所以也不能说pluripotency gene在的时候lineage
marker就不表达。

The stories of all of our paternal lines can be traced back over 275,000
years to just one man: the common ancestor of haplogroup A. Current evidence
suggests he was one of thousands of men who lived in eastern Africa at the
time. However, while his male-line descendants passed down their Y
chromosomes generation after generation, the lineages from the other men

it could be quite difficult to distinguish these two in some patients.
basically, in AA, blood stem cells are decreased in number but their
functions are normal; in MDS, the number can be either normal or decreased,
but their functions are abnormal. as a result, in AA, the morphology of
peripheral blood cells is normal; in MDS, some cells have abnormal
morphology and may be destroyed prematurely.
