你对Boltzman's principle的理解完全错误的。Boltzman's principle 只能用来描述
类似于 气体分子/离子 这样的微观分子,而voltage sensor简直是不知所云。
(1)在原文当中,已经很明确的写了,“From Boltzmann's principle the
proportion Pi of the molecules on the inside of the membrane is related to
the proportion on the outside, PO, by Pi/PO==exp[(w + zeE)/kT]," 这是非常经
典的热力学上描述分子在某个能量状态下的概率。比如说 “Pi” 就是 某(带电)分

键的是switchable dye. 只要掌握了这个东西,STORM也好,PALM也好,高中生、甚至
初中生都可以理解原理,甚至原创出来。庄小威是化学出身,之前研究的一个方向也正
是这个switchable dye, 比如她在2005年的这篇论文(STORM, PALM都是06年的工作):

其实楼主的问题我以前问过,当时没有什么讨论。再跟进一点想法:
高通量蛋白分析可以分in vivo(proteins in natural environment)和in vitro方法
。in vitro主要是protein array和Y2H,in vivo当然是MS。不用多说,各有明显优缺
点。
NGS的优点在于它是separation independent & single-molecule based。其他的
single-molecule技术都不能做high-throughput,原因是受限于limited fluorophores
。同理,蛋白分析能够也做到SM sensitivity和separation independent,将是true
breakthrough。
MS的优势很明显,但是sensitivity, throughput和cost-effectiveness都有瓶颈的。
所以in vivo基本无法做到single-molecule proteomics。但是对于in vitro,如果

BRCA is not used for screening for breast cancer or ovarian cancer in
general population. For breast cancer screening, still mammography for older
than 50 yo. For ovarian cancer, no screening markers. BRCA test is only
used when there is strong family history of breast/ovanrian cancers.

I do not know.
What I am thinking now is, since in liquid, the distribution of molecules
is random, therefore, the "packing" of molecules should not be a factor.
Therefore, this leads to the assumption that the absorption wavelength of
liquids comes individual molecules.
However, what lets the individual molecule have color preference? Why
the absorption wavelength of metals is not governed by this molecular
absorption preference?

Single molecular biophysics这个领域不能算Steve Chu推动的吧,80年代到90年代中
期就有一些组用STM, AFM研究单个生物分子,Ronald Vale, Steve Block80年代末90年
代初就在玩 single molecule biophysics了,更不用说70年代就发明了patch clamp,
也算single molecule biophysics啊.我觉得Chu只是在适当的时候说了些话好像就把这
个single molecule biophysics变成显学了,其实已经有很多人在做了。
其实对于现在这么多人做single molecule,我倒是觉得不一定是什么好事,虽然这个蛋
糕很大。

Man, it seems there are two issues here:
(1) Whether a molecule can absorb photon and have direct vibrational
transitions.
(2) If the above is true, whether the transition is in the IR spectral range
or not.
Your book answers the second one. On the first one, it depends on whether a
particular mode of vibration changes dipole moment or not. All polar
molecules are IR-active. But the reverse is not true since non-polar
molecules can have IR active modes, too.
