底敲除（把卵子mRNA也去掉），基本上什么都发育不出来，流产都免了。在果蝇胚胎发
育时大部分细胞都会变成神经细胞，是CPU级别的pathway。Notch pathway在成人中的
功能不完全清楚，但已经知道的部分也相当重要，比如免疫系统的维持。所以针对r-
secretae的毒性估计就从此而来。

1. 清除Abeta的药物（免疫方法或Bace1抑制剂）都已经出来，虽可以清除Abeta，但根
本就没有改善dementia的疗效。所以这样的研究不但临床应用可能性很低，而且又没有
疗效，别人觉得这样的研究毫无意义。
2. 没有一个老鼠模型是真正的Alzheimer。纯粹的Abeta高表达和形成plaque不导致
neuronal死亡，除非同时做Tau mutation。 这跟人为的其他方法杀死神经细胞没有什
么区别，所以和alzheimer就没什么关系。而且，tangle被怀疑只是neuron stress后期
的东西，不在早期起作用。就是说在AD被人脑内杀死neuron的根本就不是plaque和tau
的直接作用。所以，用所谓的AD老鼠模型其来做疗效只能是相当于体外模型，没有强有
力的说服力的。
早期清除Abeta肯定可以防止Alzheimer，这已是公认的事实。但疾病进程一旦启动，很
