so, for trouble shooting:
1. run gel to check your sample before digestion but after
purification.
OK --> digestion problem
smear --> purification problem.
2. if purification problem, try to use a PCR purification kit.
3. if digestion problem.
A. make sure the fragment you cloned did not contain any
HindIII/EcoRI
B. try a sequencial digestion..

首先，offer letter是四月初发的，很正规的written form,但是当时HR没有提及任何
关于export license的细节。接收offer之后一直跟公司有联系，background check也
没有说有问题，上周supervisor和同事还发信在帮我找房子。完全没有悲剧的征兆...
结果昨天下午５点多接到HR来信说要withdraw,整个人shock到了.....所以以我的亲身

platform by scientists in Amgen, Biogen Idec, Genetech and GE, the best guy
in this group is my best friend (a consulting guy with big name in protein

Dynabeads和他们的mRNA purification kit。 按照他们的protocol做完后跑胶发现里
面还是有挺多rRNA。上网搜了一下发现有的人建议连续做2次protocol里说的
purification。 请问大家有什么建议么？
我试了一下发现第1次纯化后获得了大概3.8%的total RNA。第二次以后有1.5%的total
RNA。我不太确定这样连续2次purification会不会损失太多mRNA。 |

gel purification后的dna，260／230很低很正常，可能和残留试剂比较多有关。只要
dna浓度可以，不会影响后续步骤。
不过gel purification会导致TA overhang丢失，连接效率降低，可以试试pcr
purification后直接连。 |

发帖数: 64 | 26 gel purification没那么容易引入mutation的，只要你不用紫外使命的照。我做过的克
隆多到没法数，从来都做gel purification，2k以下的片段，我最多挑4个克隆去测序
，大部分时候是100%正确，还从来没遇到拿不到正确克隆的时候，而且做了gel
purification，会给你后来的几步省去很多麻烦。
照胶的时候放个尺子在你的gel旁边，这样你就知道该往哪割了。我用紫外最多不超过
30s。你要担心胶纯化后7k片段浓度太低，同时做几管PCR，全部跑胶。

然后平常买菜放东西等等都在尾舱,有时车上还会剩点儿开盖的零食比如烤鱼片儿
结果就是时间久了车里有异味,而且送去做detailing清洁完之后还会有

用户都反映几乎没有作用

点进去看了一下review,很多评论,而且一边倒的说效果显著,唯一可能负面的
就是说噪音有点儿大
看完review就决定买了,当时被异味困扰实在太厉害
这个东西内部有个主动的抽气风扇把空气吸进来过滤,
然后设置了三级filtering process:
