基因多长？
我用的RLM-RACE，就是RNA ligase mediated RACE
你搜文献应该能搜到
具体就是利用RNA ligase可以连ssRNA+ssDNA
合成DNA oligomer，上面有一段adaptor sequence（跟你的目的基因/目的基因组没有
同源性就行），5’端和3’端都要-OH吧，然后RNA ligase会催化把这段DNA连到mRNA上
，再做1st strand cDNA synthesis
然后用gene specific reverse primer和adaptor forward primer (可以是你前面用的
adaptor，也可以重新设计引物，往里面来一点，做巢式PCR）来扩你要的基因
这样子好像应该比$740便宜很多 |

看了一下RLM-RACE KIT的PROTOCAL，它好像并没有对3'RACE做出什么改进噢。貌似是针
对获得5'端序列的，或者针对扩增全长CDS的。而且大家貌似普片都认为3'RACE比5'
RACE简单容易，为啥我就做不出来呢。。。。
一直在用Roche的5/3RACE kit，也试过用GC RICH KIT （老板说AT RICH的SEQUENCE也
可以用GC RICH KIT，但是我没有做出来）。 |

今天终于跑了很早之前报的一个local 5K race。为了这个race，给自己制定了一个10
周的训练计划，从8月中开始准备，可惜训练计划刚过半就半途而废了。。。原因有很
多，起始于训练刚过半的时候打鸡血的跑了个10K，把自己跑伤了，于是歇了一周，从
那以后就开始彻底打酱油，每周跑1－2次。。。比赛前一周索性一次都没跑，心想着5K
反正跑不死，最多慢点，今天没啥压力就去跑了。
最终成绩是27：34，我自己的表测的，官方成绩除去出发的那点delay估计能再少点。
反正一上来就是跟着人群跑，pace 8：30左右，我之前从来没有在这个pace上hold住
3mile，甚至没有过1.5mile的。心想着管他呢，豁出去了！就这么不知不觉跑到中途折
返，开始觉得有些累了，很多人开始超过我，我小心翼翼的保持住速度。2.5mile过后

刚跑完thanksgiving gobble gobble 4 mile race @ davis square，somerville MA，
用了31min34sec。 比我的目标提前了2min26sec，算是超额完成任务了。之前训练只有
一次是跑到34min以内的，还是run with the wind。心情大好啊，下午可以开开心心的
是火鸡了。
今天天气真是好，阳光灿烂，一点风没有，这在11月的麻州真是是万分的难得啊。
昨晚太兴奋了，大概只睡着了5个小时，今天7点就起来了，一看外面温度不是太低，风
也不大，心情大好，吃了一片全麦面包喝了半杯牛奶就出发了。来到davis squre，注
册领T恤都很方便，很快就完成了，大概有2000人参加这个race吧。

sleep deprived -- had total about 6 hours of sleep since Friday afternoon.
went out and did an Olympic distance tri this morning -- 1.5k swim, 40k bike
, and 10k run.
Not as bad as I expected. Overall time: 2h38. Swim leg: about 31min;

age group第四（到底不是running club的，成绩都好烂啊）
跑的是和WIL512一样的race。
跑这个也算是临时起意。其实对cives大哥说的oakland的race很感兴趣，但是一怕早起折腾孩子；二怕自己跑太烂丢人。后来发现家门口就有一个race，周五联系了babysitter，基本就决定去了。
周六本来打算早点休息，但是边加班边看片，还是到了平时睡觉的点，2点半。也许多少还是有点期待，竟然咳嗽到三点多都没法入睡。（白天不怎么咳，晚上咳的厉害，是不是什么百日咳？）
家门口比赛就是好，8:30的比赛，7:45起床，8:00已经把面包塞到嘴里，把娃塞到车上，8:10不到已经到了比赛地点了。一边到register的桌子那里现场报名，一边跟babysitter联系接头。
停车时，看到无数跑步身材的人，心里越发萎缩的惭愧，也没好意思热身（本来也没这习惯）。

8:40 5K集合，发现好身材的人并不多，估计都按性价比跑10K去了。嘿嘿。按速度站队，前排好多小娃娃，当说站前排的是pace6以内的 |

结束了一周的连雨天，saint louis终于迎来了艳阳高照。气温42度，微风，race上午9
点开始，正适合奔跑。这次race的
人很少，大多是walker，runner估计不到100人。course很平。
出发之后我紧跟着第一集团，305也派上了用场。出发时的pace大约为6'30''，慢慢提
速了一点。过了1.5 miles，第一名
脱颖而出，我紧跟着第一名脱离第一集团。第一名也戴着305，pace很稳定。跑到2.5
miles的时候我的呼吸有点散乱，左
下腹也开始有点作痛，被第一名慢慢拉开距离。最后冲刺的时候已经发不上多少力，只
是“勉力”冲过终点。
问题：我每次tempo跑的时候到第3miles左右都会出现左腹痛的情况，肌肉感觉还可以
发力，但稍一用力感觉里面震的很

几个星期前想跑race, 在running in the usa上这个race还没有出现，所以就注册了上周跑的那个. 后来这个race出现在manual上了，名字很好玩，叫“world famous pig run”，就在我们村down town.我一下子就被吸引了。另外，跑版大牛们看完report说我上次没有all out, 我想知道自己全程all out能跑多少。
还是下午，7:00pm. 天真热啊。跟BF一起到比赛地点的时候差不多6:30pm, 交钱注册，拿号码布。上次跑前没有喝水，这次去了趟洗手间后就一直喝水，生怕中途渴了。观察了一下，这次好像有一两百人的样子，大多数是壮年，20-45岁。
不罗嗦了，我早早的等在起跑线上。旁边一个是巨fit的小年轻，20出头，6 pack 清晰可见 （他后来是man third）。同时我旁边后面也差不多都是很嫩的年轻小伙子。（真的不该跟他们站一起）。开跑的信号竟然是alarm, 不是枪声。alarm一响起来小伙子们就冲出去了，天啊，太快了，我也不由自主跟他们冲起来，还是脚尖着地！真是一个big |

加州一个叫Gordy Ainsleigh的男人本来要参加horse race, 但是他的马在比赛前瘸了
，所以他就用自己的脚去跑。汗啊，中间还在医疗站让兽医检查了一下身体，最后在24
小时cutoff前17分钟跑完。后来越来越多的人用自己的脚来跑这个horse race, 终于在
1977年runner太多把race horse挤掉，正式有了一个100迈的footrace. 太彪悍了！ |

10k的是downtown的Susan G. Komen - Race for the Cure. 不是不想支持fighting
against the breast cancer, 而且也正好要去城里办点事. 可是一想到要跑完汗水淋
淋熬一天, 就犹豫了. 且race day registration又要起的更早, 自己又在和感冒交战
的最后关头. 还是偷懒点, 到附近的树林跑吧. 对, 就是已经跑过两次, 写过两次
report的Busse Woods. 前两次都是跑一个loop, 今天是只跑10k, 目标争取40分.
今天天气有点凉, 大概53F. 磨磨蹭蹭的, 开始跑已经8点多了.

德国人民，特别是柏林人民，看起来酷酷的，但是都很有礼貌很有涵养，做事情非常认
真到位，非常的喜欢
赶到比赛地方找存衣服的地方，发现Female的都在离起点比较近的地方，很好
存包的时候和一个德国高妹搭讪
她说这是第二次跑了，第一次是7年前，跑了4小时50几分
老公在美国工作，为了支持她跑步，专门回来几天帮忙看两个孩子
她才可以有时间去柏林跑步
可惜感冒了，so she'll run as much as she could, and let the endorphin do the
rest of the work!
我俩一直走到起点，高妹等我pee pee，然后一起等着开始race，聊得很开心
等pee pee的时候我后边是个scotland的mm，说8月份受伤了，但是鸡血一上来，还是很
鸡冻，据她说明年会去跑纽约，自己这次的目标是5小时内完成。当听到几个mm和自己
的target一样时候，感觉非常得开心，觉得就是run through the race, and enjoy it!

比赛的前三迈之后，我觉得not my day，接下来就是或早或晚的问题。前半程not bad，但是一不留神就会slow down，而不是芝加哥时一不留神就跑快。前半程有至少有两个大坡，爬完之后心跳就急剧上升。过15K的时候1:09，半程的时候1:37，和芝加哥的时候持平，不能说不好。但是我自己知道，今天不可能像在芝加哥那样finish strong。原因很多，最简单也最致命的一个就是训练荒废了三周，肌肉的承受能力立刻就下降了，归根结底还是实力未到。一些客观因素，岔气、calf开始痛、脱水等等，就都是表象了。

before a marathon race is definitely bad. Have a good recovery and come back
strong for next year!

set your PR. And the support during the race is not great either, at least
they should have more water stations and serve bottle water.
If I am not doing NYCM next year, I will run the CCM again I think. I like
local races. If someone is able to run with 2:45, then there is a chance to
get the prize money:)
