I'm a tanker. 50% of my playing time is either in tank or IFV.
Now before the patch, Jets/helis are minor headaches. I've gotten taken out
by jets in one round of attack very rarely. I've been taken out by namely
the A-10 so quickly that I can't react, maybe a few times only. (I've played
~1000 rounds of BF3).
I judge, by the rate of decrease in my tank's health, what to do to avoid
being killed.

升级，合成。直到最高级。 非常耗钱。。
一般流行 4-7，4张卡片， 7次升级 就是4张卡都满级在Envolve, 每次只envolve满级
的卡。 96% efficiency 相对却便宜很多。

如果rare的是体力卡那就更牛逼了，能卖1k5～

度假回来用大伊布踢了将近20场，有点小失望。可能13的时候太bug了，心理有落差。
现在场均不足1球，表现差强人意。可能跟我用的4141阵形也有关。现在依旧在D4挣扎
。出了patch之后，finesse shot威力大减，大伊布的威力也受到了影响。
这次UT前后开了2W点，之前将近1W点运气比较差。后来稍好一些。不过跟黑卡帝也是没
有办法比。我觉得我属于开卡运气比较差的了。算了一下平均卡包收益6000 coin. 比
较适合前期快速致富没时间搞trading的人。后期还是买金币更划算。

to very delicate region where you cannot apply eye cream. I feel the

排名第三的彼得堡的state hermitage museum，拍过电影russian ark，就记得金碧辉
煌，没看到什么画……
排第15的Yale Center for British Art我也常觉得比英国的画廊都好……小而精致，
各个时期和画家的代表作收集得很精，介绍也很精，看完印象很深。建筑也很宽敞明亮
，令人愉快，不过太亮是不是对画不好。
有个Doha的Museum of Islamic Art。希望有机会去。
台北故宫上榜，北京故宫没上，倒是北京的789工厂榜上有名。说最近有一个Duchamp
and/or/in China的展览，到六月中截止。听着挺有趣，不知道goldy去看过没。
第50名是波士顿的museum of bad art，听着很好玩，有人去过吗？
Times另外还评选了世界50 museums，我没Times帐号，看不到全文（这篇50 galleries
是其他报纸转载的），谁有帐号的能不能分享一下？谢谢！

学术上，现在普遍用NGS来study家族遗传病或者sporadic genetic conditions。以最
近发现了很多新的基因（Autism，RP等）理论基础是（Rare disease, Rare variants;
和 common disease, rare variants）不同于GWAS的（common disease common
variants）现在看来比GWAS和linkage更易于发现disease allele。感觉几年内还会有
很多的新基因被发现。但是技术是比较成熟的，我感觉应用的关键是对基因功能的研究。
Clinical上，有几个方面： 1）产前的诊断：尤其是现在non-invasive的手段，可以在
母亲血液中分离胎儿DNA，通过NGS诊断。不但可以检测Down syndrome之类的，还可以
检测很多单基因疾病。
2）遗传疾病的确诊。同上 3）细菌和病毒感染的诊断；4）食品和药物安全检测（如前
些日子有人发的文章检测中药制剂，直接测序就可以鉴别用了些啥材料）等等。。。
美国很多人都认为不就的将来所有新生儿都会被直接测序，这必然是一个发展趋势。这

1.如何定义“遗传因素比较大”？ 想确定到底是familial还是sporadic，肯定要收集
足够的sample吧。。有时候对于有的疾病sample都很难收集。这不仅是人力，精力，
funding的问题，有的疾病病人死的很快，或者sample本身很少，总是很难碰到一个好
的大的pedigree的
2.NGS技术本身我当然是很看好的了，肯定越来越精确成熟，而且越来越便宜。但到底
能有多便宜？什么时候可以很轻松的给每个人做全基因组测序？read length可以达到
长?（肯定是越长越好）计算机的硬件能跟上NGS数据发展的趋势吗？
3.生物信息分析。我只能说现在的bioinformatic pipeline，除了read alignment和
SNP calling变的非常成熟（不仅sensitivity/specificity很高，而且可以做
population-level的分析），在其他方面，要么很艰难，要么很混乱。就是说无法达成
一个统一大家公认的最好的pipeline，我开发一个方法，你开发一个软件，最后把使用

“4.对基因组，genome biology本身的认识。其实纵然是whole-genome sequencing，我
看大部分的paper也就是先找找coding region/splicing的SNP/indel；或者说大家还是
gene-centric的。现在我们对基因组的认识还很不完善，且不说loci interaction，就
连最基本的每个loci是什么作用都不清楚。很多基因的功能都不知道，而且gene本身的
概念也在被扩充，不断有新的gene被发现，以及各种新型的什么miRNA gene啊，
linkRNAgene被发现。还有很多regulatory region比如被ENCODE给annotate出来。除了
非常明显的罕见的large deletion/duplication，或者一些repeat expansion，我们的
精力还是停留在missense mutation上，因为这个最好解释。而splicing site，或者
regulatory region有一个哪怕是罕见的A到T的突变，请问你能立刻给我解释下这个

发帖数: 2518 | 47 貌似你说的coverage是，比如你SNP calling一共找到了多少SNP
call出来的SNP越多，那么false positive的几率就越高
然后我们附加一个additional filtering，可能SNP总量就少很多，增加了可信度，但
同时may lose some power(sensitivity)
这个我很赞同。
但很多的project，我们要找的是rare SNP； 对于那些很真的rare SNP，不管你如何调
节这个“coverage”，those rare stuff are always at the top of the list.
另外我很赞同的是，未来computer硬件是个大问题。人类基因组太大，TB级别的
cluster已经不够用了，或者用起来很痛苦

里寄好？离大城市太远了，不能去游行示威，只能写点东西尽一点微薄之力。信写的可
