在最近一个访谈中，２００７诺奖获得者Martin Evans（由早期对胚胎干细胞以及转基
因老鼠的工作而获奖）对最近的两篇被retract的 Nature 上的干细胞文章（一篇是关
于物理压力，另外一篇是关于酸液）直言不谓的对Nature的editoral process表示不满
。并呼吁年轻科学工作者们不要全部相信文章上的所有说法而是要仔细看其中的数据并
做自己的独立判断。

1） 这个我昨天看到时候也很激动，但是仔细考虑了下，它的应用比CAS9的路障估计要
多太多，正是因为它太优美了，需要的东西太少，那么从临床治疗角度它可能出现的胞
内的ssDNA会大规模增加它可能的off target---所以它的优势潜在地是它致命的劣势，
这个不确定性对于临床转化是致命的；但作为研究工具，如果果真像文章所言，那么我
觉得很快如果不取代Cas9，至少也会同CAs9平分天下；
关于这个新技术，我现在已经有N多的想法做什么了，但是需要人手呀！！！而且必
须快手，当年Douna文章一出来（不是张峰的文章出来），我就设计了7个课题，两个我
们还在干,一个我们投入最大的，后来发现理论上非常牛，非常好，现实操作上可行性
是无，光大提的试剂盒估计得用至少4000个以上：）））（不过我们还在想替代的办法
，日本人去年的一篇文章让我对这个课题又有了重开的兴趣，不过需要强大的技术骨干
才行）， 其他都眼睁睁看别人发CNS....
2）第二iPS获诺奖是必然而且是well deserve,尽管我从来都没有觉得IPS可以走通临床

iPS本身最大的创新可能是1）技术的简易化，使得其迅速普及；2）其本身机制的研究
促进了epigenetics等领域的发展。
reprogramming这个概念早就存在。就体细胞向ESC逆编程而言，SCNT应该比iPS更好，
可惜的是，到目前为止还无法在人的细胞上完全实现。
iPS细胞刚出来时，几乎所有人都对其临床应用充满了期待，随着研究的深入，该领域
内怀疑其clinical therapy价值的人渐渐增多，其主要弱点有：
1）iPS与ESC的identity：Yamanaka现在似乎仍然坚持iPS与ESC是非常相似的，但是他
本人也承认现在iPS还根本无法取代ESC;相反的一派认为iPS与ESC根本就是两种不同的
细胞，无论是在transcriptome水平，还是epigenetic水平——蛋白组水平的研究，我

我这车可以手动关棚，但是据说手动关了之后，需要reprogram，否则电动会乱套。而
reprogram又非到dealer不可

200刀铺子搞得,没有reprogram. youtube有不reprogram的搞法,要有备用电源.我也不
识狠明白为何有时候必须reprogrem,感觉很危险, 又一次我清电脑没弄好,弄出德文来
了吓够呛, firmware还是不要乱刷.
自己搞估计也要120左右,懒得折腾
bmw的电池电量比较大,因为停车也很多东西在消耗电能,最好买大家用了都说好的
个别人说walmart买了也可以, 你可以试试
这玩意没几个钱我德观点不省买好的自己换找铺子都可以我的电池是第七年敲的翘之前
有症状打火咳嗽然后就不咳嗽了死了各种警告阿吓死我了 bmw的工程师应该拉出去枪毙

这个主要是优化PCM（或BCM)里面的参数和程序设置，据说可以有效改善车的运行状况
，比如transmission会更平滑，MPG会更高，可以提高扭矩。 |

发帖数: 161 | 42 我的01年的century，在我手上3年开了快4万迈，就一个买菜车，开过1000迈左右的长
途4次，主要Local跑。别说这个老车，虽然加速肉点，但是乘坐很舒适，自动大灯，自
动门锁，拉手刹挂档报警等等自动功能，还是有不少人性化的地方的。
修过如下东西：
1，驾驶员侧门上的开关，自己换的，40左右？不记得具体数了
2，前面板的空调开关module，100多点， 自己换
3，驾驶员侧前门和副驾侧后门的regulator，花了180，自己换
4，换了一个遮阳板，dealer处换的，花了150，可能稍微多点，应该少于200
5，check engine灯亮，查code，说是EGR valve问题，自己换了一个，花了70，故障依
旧，找dealer，reprogram，花了99。
6，换了3个尾灯灯泡，花了3刀
7，换了一个电池，80
至于换轮胎，换油就不统计在内了。
算了一下，竟然花了我差不多800大洋，这也太不reliable了...............
看看NBHH的DFBB，人家从来都不用换东西，机油都是2年一换，最终卖出去还能挣钱，
长叹一声，不能比啊。 |

第一辆2006年91222mile，车主为一年轻女性，丈夫在volkswagon的Dealership工作，
自己对车作了9万Mile的大整修，更换了timingbelt, water pump, brake等，车有一处
小擦伤，历史干净，车主说后保险杠曾经伤到，更换过，没有出过其它问题.autocheck
评分91，售价10500
第二辆车2007年89233mile，车主为中年男子，开长途出去工作，车有定0期保养记录，
车体和内部都比前一辆干净。但没有做过9万mile的大维护。autocheck评分90，但
carfax上曾经有过一次召回记录，ECM reprogramming.没有事故。价格11800
两辆车型号相同，年份差一年，其它历史几乎相似，不知应该怎么选择？

发帖数: 5998 | 49 I am pretty sure you can add back up camera to your OEM head unit. I am
about doing this to my 2009 ML. All I need to do is to install a camera,
which you can get from 50 bucks to about 200 bucks depends on what brand you
want to, then go to a dealer to have the head unit reprogrammed. In my case
, a MB dealer charges 130 bucks to turn on the backup camera function in the
COMMAND. So, I can get backup camera when reverse is engaged and displayed
in the OEM screen for under 200 bucks.
