around CDS starting point.
Now, I have found the Tranlation Start Site, and also I can make sure the
downstream coding sequences after the TSS. So now I need a comfirmed 5'UTR
sequence, at least some of the sequence near the TSS. Do you think the
unstream of the TSS in the gemome is 5'UTR ? or at least we can make sure of
a samll seuquence before the TSS?

cloning primer,发现有两个看起来都可信的reference sequence,blast了一下,发
现这两个sequence在CDS区有一部分是相同并连贯的,其它就都不同了。其中sequence
1只是cds,并且是partial cds,没有stop codon;sequence 2是complete cds,包括
非CDS区,好像是由NIH MGC (Mammalian Gene Collection) Project来的,但由
sequence 1衍生出的AA序列却比sequence 2衍生的AA序列还要多50多个AA。
我选择了全长的sequence 2设计primer,一共设计了3对,PCR后都未能得到预期bp的
band(虽然有的PCR出来是single band),我每对primer都挑了最接近预期长度的PCR
product去sequence,但都不是我想要的gene。。。难道是sequence 2本身就有问题么

首先观察可得,对于所有相同长度 sequence (前面的 pair),只要保存 last number
最小的那个就行了,因为其他的都已经是 dynamic programming 的 decision tree 里
面的 suboptimal subtree,整个 subtree 可以一刀直接 prune 掉。
同理再次观察可得,对于所有相同 last number 的 sequence,只要保存长度最长的那
个就行了。。。这个及以上那条可以得出 ordered 属性,也就是对于保存下来(没有
被这两条规则 prune 掉的)的 sequence,sequence 长度越长,last number 必然越

主要是in vitro 的protein实验。我们鉴定了很多新的splicing isoform,其中有不少
有premature stop codon(可以是splicing 造成的,也可以使indel造成的)
现在我们想做一些quality control,去掉一些不靠谱的transcripts,于是出现了分歧
组里的大姐想法是要尽量跟已知的protein一致。把每条isoform对应的protein align
到已有的protein sequence上。如果整条protein基本align上去,即便使truncated
protein, 不管什么原因,都可以作为partial protein 保留。但是如果shifted frame
的amino acid sequence长到一定程度,那就认为这些protein sequence跟已有的
sequence太不一样,要除去
我完全明白大姐为什么那么想,因为我们实际test的是一些protein sequences

sample
当然我的问题和你的不同,因为这里的screen hundreds samples基本上还是用sanger
sequencing去测某几个位点(当然也在策划deep sequencing);而且问题是ALS属于
complex disease,找到的基因都只可能是account for 1% of all samples
所以我好奇的是你这种疾病是不是monogenic disease;如果是的话就相对好办吧,哪
怕你只有三个sample,但assumption是所有的病人的致病机理是一样的,你先做着这三
个sample也总是可以发现点规律。当然我不知道甲基化,乙酰化的deep sequencing的
成本,但我相信总归算target sequencing,算算exome sequencing现在的成本降低的
厉害,所以我猜想你的这个也不会太贵吧?
做这种研究的思路我觉得一般都是用deep sequencing测序一部分的重要的样本,找到

Hi,
I tried a couple of transfections with the NgAgo plasmid from Addgene and
the 5'P oligo against GFP from the paper, but I did not see a dramatic
reduction in GFP+ as seen using Cas9 and a GFP sgRNA.
Anybody tried as well and came to the same (dead) end?

不能生产激素或用一个小分子化合物抢占激素受体上激素的结合部位。但多年治疗后,
会有相当比例的病人产生抗药性。这个抗药性机理相当复杂。我们实验室一直致力于筛
选和现有药物不同的机理的小分子化合物,包括受体和DNA的结合或其他与受体相关的
pathway的inhibitors。前几年成功地筛选出了几个不同机理的化合物,个别在细胞水
平达到了nM的级别,动物测试也在10 mg/kg的水平。
最近在这个领域对抗药性肿瘤的研究发现,部分病人的抗药性是因为受体激素结合部位
发生了突变。一个或几个氨基酸的突变就能导致肿瘤细胞的生长不能被现有药物抑制。
所以,我们很想知道我们筛选出来的化合物对这些突变受体是不是也起作用。理论上来

打赢的,一定要粉碎美国人阻挡咱们大国崛起的阴谋的。当美国提出500亿的清单时,
咱们针锋相对;当美国提出1000亿的清单时,咱们继续应战;而当美国把清单的总价提
高到2000亿的时候,一夜之间,美国就不是阻挡咱们大国崛起,而是有利于咱们大国崛
