staining 那个蛋白居然还在... 请问会是些什么原因呢 我的mating scheme如下
Cre+ ; fl/+ male x fl/fl female 得到 Cre+ fl/fl female
现在想到的问题有一下几个 1.可能蛋白在Cre前就已经表达了
2. 同时excise 两个有loxP的allele是不是不够efficient阿? 由于我做的是oocyte 所
以尽量选择让公老鼠carry Cre, 因为是homozygous lethal 我不是不是需要一步一步
的去除基因呢
请问大家我该如何着手解决这个问题? 谢谢了! |

1. 我用的是ZP3-Cre
2. 做western有点不太现实, 蛋白表达度很低, 而且卵子那么小... 需要至少250以上
的卵细胞才能detectable, 需要很多的老鼠, 目前没有那么多.
3.恩 打算下步试试这个RT, 请问从zp3-cre从表达到我做实验应该至少有25+的时间
这个蛋白稳定性很好?
4. 这个应该没有错把... 不然太....耸了...pool了3只老鼠的卵子染色的 每个都有
specific staining pattern 跟control一样...

Chromatography A.
One of the reviewers gave the comments:
If the manuscript should be published in an analytical journal some
modifications have to be done. Sensitivity, specificity and accuracy are not
used as typical in analytical terms. Please rephrase.
I'm clueless about what the reviewer meant to say. |

letter of interest and intent directly send to them. Can some one provide
some clue about this?
p.s. I already received a rejection letter from this program a couple of
days ago, both in myERAS and my e-mail box. I guess they are just sending
out the request to all applicants including those they already rejected. I
am afraid even if I reply, they won't touch my profile anymore.

和医生吧，我觉得不能他们要什么就给他们什么，要跟他们讲道理。实在讲不过，把他
们要的东西给了，也得把这个东西的局限性反复说清楚。不过统计师和医生的交流难度
好像是个永恒的问题 :-p
关于诊断这个，和医生的一个交流办法是，问他们，愿意用多少个false positive换多
一个true positive。比如癌症，如果要开刀诊断，有的情况下医生就会觉得，白开30
个病人得到1个true positive他们就很满意。如果手术伤害很大的情况，可能医生就会
觉得，白开10个病人得到1个true positive他们就会满意。这样再换算出一个评价
specificity和sensitivity的公式来。 |
