H1B回北京签证，已经定了回美机票，但是这个如果填yes,表上还会要求你填一个date
of departure from US。这怎么填好啊？是填个估计下回离开美国的时间？
还是干脆选no好，这样只用填个入美时间，然后根据H1B的有效期写length of stay就
可以了？

感觉两个我们都能填，因为网上两种都说了如果不确定日期，可以填估计日期。
填哪种麻烦少点，签过可能性高点，还是没影响？
另外，选签证类型，应该是business or tourism (B1/B2), 还是tourism/medical
treatment (B2) ? 我父母是tourism， 但不是来看病的。

似的.
2.不能太specific,否则你就等着audit吧.从没听说过可以要求会议上发表过论文的,这
是unusual requirement,必定audit.如果不是非常明显的(比如申请中文口译的perm),
不可以要求中文,否则必然audit.不可以要求在微软工作过,这是unusual requirement.
prefer可以写任何东西,但是这是没用的,因为你不能因为别人没有prefer的东西就拒掉
别人的申请.工作年限当然是可以和你的符合的,只要不是看上去太离谱的,应该问题不
大.publish app这种东西也是unusual requirement.
反正基本上就是一个学位和一个工作年限.别的加了是自己搞笑自己.

把我工作中用的软件都写上去了,写的很具体.因为我的广告的job description 就是这
样写的(工作需要的skill). 我会不会被audit?因为写的太多太具体.DOL think it is
too "restrictive"?
有些人说有些skill如果能在reasonable time 学会就不能要求申请人在申请时已具备
这一skill. But what's reasonable time?

query在Java程序里操纵数据库，那些java 程序引发的编辑操作（update，delete...)
会不会也被log。

security sector,我发现SQl Server Express 有basic auditing 的功能，但是没有
fine auditing的功能。但是我怎么就找不到在那里设这个database audit
specification. 我用的是SQL Server 2008 R2. 是不是版本不对。
另外，如果不费您太多时间的话，就把我第二个问题也一并回答了吧。我现在就是在用
JDBC编一个小java程序来管理一个很小的数据库。有那么几个人用这个数据库。就是希
望能够log一下数据库编辑的记录来防止别人干了“坏事”后赖账。用户是通过Java程
序support的界面来用数据库的。 |

出来。请问大家有什么推荐的抗体吗？像anti-phosphotyrosine啊、anti-
posphothreonine这种的。不知大家觉得有没有其它更好的办法可以监测或定性这类磷
酸化作用？

确实这个问题挺困难，目前还没有什么号的方法。我很头疼，难道我只能靠猜测几个
potential TFs, 然后做ChIP-seq了 （对于我的project）？这样发现某些新东西的可
能性就不大了。各位说的都很有道理：
Shakuras说的DNA长度的问题，目前很多人做的（Bulyk @ Harvard）都只是10-mer,至
于想模拟chromatin之类那么长的DNA我觉得很难。
难道是个悲剧。。。。这个project挺有意思的，以前发过贴问过版上各位可惜没人回
复。
即将拿到ChIP-seq的数据，但是不知道下一步怎么走。如各位说的：
怎样Purify specific的TFs complex，太难了。

这个想法我在2005年1月之前，换实验室时，和PI说了，他听不懂这张PPT意味什么。
后来我又换了实验室，2007年（2008？）某天新老板和我听完某个不相关的seminar，
和我说起这个，他才开口说了2句，我就知道他想说什么，
他的反应是，i have not finished my briefing yet, you really understand my
point? 于是就和他说，of course i know what you want to do,然后讨论了半天，原
来他的朋友有这个想法 etc etc，后来我又做了一些Homework，还是决定放弃。这个很
不好做，估计是个死途。
这个技术最大意义是：能找到locus specific DNA-protein complexes。目前的通行做
法是，已知某个protein, 你用CHIP，证明在某个Locus上面，然后开始making story，
编剧。但其实，那个Locus上面可能有几十种特殊蛋白在调控！！！
