中最重要的一个概念是近系小鼠（inbred strain）。Inbred strain 的特点是所有的基
因位点都是纯合的（homozygous）。严格的定义是经过连续二十代兄弟／姐妹交配（
brother sister crossing）的小鼠品系。由于是兄弟／姐妹交配，一旦交配的雌鼠和雄

抽质粒用的是Qiagen mini-prep kit
跑电泳发现是很亮的smear

没有细胞毒性。用的培养基是LB+M9 salts，抗生素是Amp+Chloramphenicol。关键是以
前摇菌（同一个strain）都没有这个问题，最近屡屡出现，我不知道是哪里
contaminate了，如果是phage的话，就恐慌了，据说很难清除phage。 |

For the genes involved in TCA cycle, it really depends on the enzymes.
Sometimes there are alternative pathways and metabolic flexibility. There
might also be other isoenzymes. You may add some substance to the media or
clone the similar genes with the same function from other bacteria to
restore the phenotype. |

比如这样一个
XXX-rtTA;tetO-Cre；XXXfl/fl
虽然是triple transgenic..但是理论讲，在没有诱导
前是没有phenotype, 完全vital的。所以一旦拿到了triple transgenic strain就可以
拿非诱导的mice来keep line以及generate new pups.效率会高很多。我现在在做的耗子就是triple transgenic.如果算上里面的R26R-LacZ reporter那其实是tetra-transgenic.
生成第一个triple transgnic strain需要大概半年时间。
做CMV-Cre全身诱导的问题一来是缺乏tissue specificity.二来那个未必在全身都会有同样的效率。我记得好像CMV-CreER在liver 和lung的效率就不好。反正实在没法了才来这个。
诱导系统目前就doxycyclin和ERT俩了。Jax lab都有。没事上JaxLab网站浏览他们的各种mouse lines很有意思的。 |

P should have same function with respect to X1-R2 and X2-R2. Once bound with
X1 or X2, both R1 and R2 should block the transcription of eGFP.
R1 and R2 recognize different inducer. The inducer for R2 is a universial
