primers for matA1, matA2 and matB1, matB2 and perform colony PCR. If it only
gives product for A or B, then it is haploid; if it gives product for both
A and B, it is diploid.

。但并没有很多stem cell 的 in vivo marker被identified出来。比如说liver
tissue,都知道其可以再生，但是它有没有stem cell仍然在争议。更莫提marker了。
endothelium是个快速turn
over的组织，但起在成体内stem cell population的identification仍然未知。
筛选stem cell marker的方式可以有两个途径
第一就是正向的从已知marker出发，比如说通过已知marker筛选出某stem cell
population, 分析其有啥gene 高表达，以此出发建立CreERT transgenic model来确认
新的marker.
对stem cell来说找到marker不是目的，更重要的是这个maker背后的mechanism. 一个
新的marker市时常可以揭示新的regulation mechanism.

首先声明一下我说的屈服强度是yield strength。
看下图，stress-strain curve总认识吧。当金属物体处于弹性区间的时候，stress跟
strain是纯线性关系（胡克定律你懂不？），也就是下图里直线段那一段。在这一段里
面，物体受力形变了，当力量松掉之后还可以回复到原来形状。
在弹性区间里面tensile strain跟tensile stress之间也是线性关系，线性的系数就是
你经常挂在嘴边的那个E（杨氏模量），它在这个区间里面是一个常量，不变的。这也
就是为什么我之前两次提醒你不要老惦记着E。
当stress超过了屈服强度yield strength（图中y轴上的sigma_y）之后，物体将进入塑
性区，这时候施加在物体上的力就算取消了，物体形变是不可以恢复的。
因此在这条stress-strain弹性区间斜率固定的前提下，谁的sigma_y（屈服强度）越低
，谁就越快进入塑性区。金属物体形变不是一个杨氏模量就能统领全局的，眼光要开阔
一些。

经常在版上看经验，受益良多，我也分享一次我面试遇到的题目，希望对一些人有用。
一个大公司的FEA职位，不方便公开具体是哪家公司。问的问题大概有：
1. 一根棍子，其中一端固定，拉伸另一端到一个strain value, fix住，然后升高温度
，再保持在这个温度一段时间。要求画出stress-time图。
2. 一根horizontal方向上的棍子，两端pin住，施加uniform vertical pressure，要
求画出momentum-position图。
3. 画出一个elastic-plastic material loading-unloading的stress-strain图
4. creep的3 regions，第二个region常用的equation。什么情况下考虑creep。
5. 描述stress relaxation

influenza 的strain经常变化，所以每年都出flu vaccine来cover当年可能流行的flu
strain.
对bacteria引起的感冒或者其他virus引起的感冒没作用
打了没坏处，只有预防的好处
而且今年的跟去年一样包含了3种strains，其中一种就是H1N1
我去年没打，因为h1n1的vaccine比较新，也没让娃打
但是去年已经测试使用了，没有听过有副作用报道

这里我就说一个缺陷
他们依赖的parent strain，就是一株，
微生物的diversity那可是相当可观（毕竟自然才是最伟大的遗传学家）
所谓同一个species里的不同isolate或者strain基因组差别能达到10%
（所以有的人还说在微生物中species概念靠不住了，炒作pan-genome概念）
于是有的基因的表型在某一个parental strain中通过knockout是永远看不出的
最简单的原因是需要一个partner，而这个partner在用这个parental strain中就没有 |

发帖数: 1007 | 47 多谢信息。我也是仔细分析了国内的做老鼠的现状，发觉确实无论时间和成本都远高于
美国，学生就不提了，而且更重要的是找不到水平相当的人可以讨论。我们这个领域国
内没人懂转基因老鼠。
我本来打算买30个strain回去，养大概100笼。结果计算下来，如果一切按规矩来，3年
下来光购买饲养老鼠就要花费150万。完全不可行。
所以我琢磨了个想法，
1。以sperm cryppreservation的方式一次性把这边拥有的老鼠通过液氮罐运回国，花
费2。5万。
2。在南京动物所复苏动物，一个strain 3800元，30个strain 就是10万左右，还算可
以。
3。复苏了的动物一方面要求动物所代为保种，一方面转运到学校的标准老鼠房，以最
低数量keep line,大概30笼吧。30个strain很多可以cross在一起，不需要每个line都
单独keep.
4.实验室里自己建立简易老鼠房用作试验，这个成本就很低了，当然就是有各种啥危险
了。我在考虑在简易老鼠房里除了空调，加湿，通风扇之外，还加一个空气过滤器，就

organ中epithelial cell的intracellular calcium level的变化。
各位的建议都是用genetically encoded calcium indicators，但查了jackson lab似
乎没有universal 表达这些indicator的strain在卖。
这就需要去买相应的conditional expressing strain再和特定Cre strain cross，需
要周期比较长。
但我现在时间比较紧张，可能来不及重新generate mouse strain，不知道有没有哪些
chemical calcium indicator能作为替代品？ |
