ER阳性是分子生物学的概念，是指病人的癌细胞表达ER。检测是antibody-based或现在
有用qPCR-based。正因为这样，有少量病人还真不能绝对说阴性或阳性。就是在阳性病
人里，也只有大约80%的人会对ER治疗的方法有效。
治疗ER阳性的病人目前是两类药物--一类是针对体内estrogen合成的抑制剂，还有一类
是针对Estrogen和ER结合的，所谓“占着茅坑不拉屎”的原理。大多数病人五年内抗药
性问题不大。五年后会有病人复发，往往是上述药物没有效果了。这个的分子机制还不
清楚。有研究ER coding region突变的，有ER高表达的等等。几个著名的Tamoxifen-
resistant乳腺癌细胞系，都是ER阳性的。
ER促进乳腺癌细胞生长的机制也不完全清楚。ER本身是一个transcription factor。解

以某个stem cell specific marker X 为例,做一个tetra-alleles model
X-CreERT;Tdtomato flox;X-rtTA;TetO-H2BGFP
在开始时候同时加以tamoxifen和Doxycyclin短暂诱导。 前两部分做lineage tracing.
后
两部分作为初始标记。H2BGFP是个long live protein .只要细胞不分裂，其能在细胞
内维持一年以上。 |

rosa26lacz, tamoxifen 诱导后24小时后，brdu feeding. 那么除了最开始被label上
的是细胞只是蓝色的外，daughter cells 从它们衍生出来的都会有 蓝色＋brdu. 不过
这里有个brdu label efficiency 的问题。
有没有一种东西弄上第一代细胞后不会随着分裂传给下一代？virus? |

Tam代谢至少要等两周才算是差不多没了，一周我记得是半衰期，但是忘了。而且如果
你代谢完之前就带出去解剖的话很可能所有的东西都要进特殊chemical垃圾，很麻烦的。
注射持续时间长的话可以考虑灌胃，不疼不痒。 |

多，否则副作用大，5天其实偏多。
还有tam必须通过肝脏变成4oh-tam才有效果，所以通过皮肤必须用4oh-tam |

细胞理论上不会丢失，所以不存在4周以后就没有了的问题。
K14-CreERT2的效率不是很高，假设你诱导敲除一半的细胞，那么另一半的细胞足以掩
盖敲除细胞的缺陷。除非你的基因特别强大。我觉的K14-CreERT2作为tracing的工具鼠
就好，要用它研究基因在成体小鼠中的功能并不是很好用，主要就是诱导效率还不够高。 |

tam注射进去就进肝脏代谢了，皮肤敲除还是通过血管到皮肤的，并不是抹在皮肤的，
不需要用4oh-tam |

发现，五年生存率都在90%以上，后期发现，五年生存率都在60%以上，早期的很多十年
生存率在80%以上。哪怕就吃tamoxifen很多也能很大延长寿命。我以前在医院，见过八
十多得的肿瘤，治疗顺利，十几年后死于其他疾病，没有复发的。

一个是乳腺癌，30岁出头，已经切除了一侧，国内医生建议吃药一年以防转移到另一侧
。大概就是抑制激素的药，国内的据说副作用比较大，推荐美国Tamoxifen Citrate
Tablets。我看了看都要处方才能买，不知道有什么办法不需要处方可以买到的。或者
其他什么我在美国可以帮忙的。
还有一个是家里老人，80多了，食道癌喷门癌，确诊了而且也转移了，手术化疗都没什
么用了，人也年纪大了。具体的病情我也不大清楚，不知道我在美国可以有什么帮忙的
，有什么药物可以帮忙维持的，或者其他一些治疗方法。
