哦~只是在cell里用的话,ms dharmacon也搞了一个。话说有个组一直在做
coadminister
ss PS ON 对其他ON的uptake有帮助,不知是不是类似原理呵呵。
degradation挺头疼。RXi那个东应该是modify过得,忘了是LNA还是啥了。
要是NP能work就好了,即使简单conjugate到surface上,从3D到2D造成的degradation
减缓就会很可观,更何况还可以包埋+controlled release,要是在加上targeted
delivery(我的最爱啊)就完美啦hahaha~说起来还是mark davis的东东最接近我的
口味啊,不知release mechanism是什么

I saw the news yesterday and have a chance to read the paper in detail
today. It was very exciting if proved, but right now I'm still not 100%
convinced for the following reasons:
1. The authors showed that cells grow better in media containing +As/-P,
than -As/-P. Could that be explained that As (40 mM Arsenate) stimulate
the cells to uptake PO4? The author never really got rid of trace amount
of PO4 in his experiment to give a clear conclusion. Also, the authors

没有很好的antibody可用。
即使有的话fractionation或者超速梯度离心也不容易区分內源的还是uptake是吧?即
使用无內源表达的细胞系的话WB的灵敏度可能也不如荧光来的高些吧。所以可能还是倾
向于找一种合适的标记物。

Medium glucose高,不代表胞内glucose高。你可以试试同时加insulin之类的处理,增
加glucose uptake,但是这样结果就难以分析了。
但我觉得细胞系在高糖下是不会凋亡的吧。体内可能不一样,我不清楚,但那也应是慢
性作用,你用细胞也试着长时间处理呢?

yes, they drained to draining LNs, I normally do tailbase s.c. the lipids
end up in inguinal nodes and axilliary nodes. And most of the macrophages (
stained positive for F4/80) are at subcapsular sinus.

这个问题也困扰我很久,albumin也可以出现在interstitial fluid里,但是浓度可能
没有在serum里高,具体有多高对我很重要,但是没有文献有小鼠的数据。

agree this, partially, and zan the upstairs suggestion on frog oocytes
1. localization
2. in vitro function, and mutagenesis
xenopus oocyte is my #1 suggestion, HEK293 , ,,,,,
radio activity labeled substrate uptake should be easier if it's neutral
transporter
NOTE electrophysiology requires special training, but it works well ,as long
as target transports ionic stuffs or is ph dependent

要开始做一个蛋白,想做it和肌肉glucose uptake相关工作,因为it有很多在神经递质
相关的文章,但查文献没有查到蛋白在肌肉中的表达,有mRNA Genechip的数据,不知
道有什么蛋白数据库可以查到相关信息。谢谢!

在贴壁细胞中siRNA了某基因,之后就有大量的碎片漂浮在medium里,镜下看就是小圆
泡,diameter < 10um,而细胞即使trypsin消化下来之后也并不是规则的圆形,这些碎
片并不能uptake trypan blue,计数活细胞(trypan blue-, diameter > 10um) 的数量
有明显减少, 但以trypan blue+ 的%算,并不比对照组减少很多,大概从95%减少到
90%
用FITC-Annexin V和PI double staining,做flow没有看到明显Annexin V+细胞群,
AnnexinV+ PI+细胞群会增加但也不会增加太多(本身%也小于10%)
用50uM Z-VAD-FMK caspase inhibitor 与siRNA共同处理细胞, 这些碎片就不会产生,
计数活细胞的数量也不会减少, 和对照细胞没有差别
请问这是否说明细胞有死亡?能否看出是何种死亡方式?谢谢大家!

1. 存在其他补偿机制的,并不只有唯一这一个transporter
2. 多肽/小分子抑制剂对HBV和bile salts的抑制活性并不等价,完全有可能设计出EC
差别很大的抑制剂。事实上,已经进入临床研究阶段的Myrcludex-B(PreS1 derived
peptide) 对HBV entry的抑制“效率”高过对bile salt uptake约2个数量级,因此在
药物正常剂量浓度下的副作用理论上不会很大。

,比如通过运动或节食。我举最简单的例子,反驳你关于基本代谢的误解,是想
stimulate你的思路,没想到你还真是算盘珠子拨一拨才会动一动。非得说FGF1没有增
加activity。
呐,我再提醒你一句,很多东西在肌肉里激活的话,都能达到insulin sensitization
的作用的,未必一定要通过增加physical activity的。exercise mimetic听说过吧?
再者,血糖如细胞便成脂肪了?没有其他通路了?
话说回来,这篇FGF1的文章里提到activity和energy expenditure (在supplemental
里),但是关于energy intake和expenditure的实验这些是不够的。为啥不够我就不科
普了,你自己琢磨吧。我再点拨你一下,去看看FGF1对肝脏的作用,加上外周glucose

Most people look at GSH depletion because the cells already have a lot of it
. Some cells if you treated them with high glutamate, it will block
cysteine uptakes and deplete its GSH level. Your probe has a Micheal's
acceptor, so I you will have toxicity around 10s microM, but it is probably
okay to do short term live cell imagining.
I wounder if you treated with certain compounds, would you be able to
determine specific GSH pool depletion because there is a cyto- and a mito-
pool of GSH.

一直好奇,谢灿的蛋白不是一个膜蛋白是怎么regulate神经元uptake钙离子的?而且,
没有Cry,MagR alone是没有磁感应能力的,Cry+MagR复合物又要光照作用下才能结合
,既然要光照,那么谢灿他们的MagR也不能实现真正意义上的non-invasive啊。
