题目:Redox-triggered intracellular dePEGylation based on diselenide-linked
polycations for DNA delivery
ABSTRACT: Extracellular stability to protect DNA against nucleases and
stimuli-triggered intracellular DNA release are key factors in designing non
-viral gene vectors. In this study, diselenide-linked polycation mPEG-SeSe-
PEI was developed as a new type of PEG-detachable gene vector for redox-
responsive gene delivery. Gel retardation assay indicated that mPEG-SeSe-PEI
exhibited good DNA binding ability above the N/P ratio of 2. The mPEG-SeSe-
PEI/DNA polyplexes had tightly packed spherical structure with a diameter of
about 70 nm and zeta-potential of around 8 mV. The PEGylated polyplexes
were stable in physiological salt condition, while fast aggregation was
observed in the presence of 10 mM DTT by TEM. Agarose gel results showed
that the polyplexes had a promising ability of efficient DNA release under
reductive condition, due to PEG detachment caused by the cleavage of
diselenide bonds. According to our previous study, PEGylated polyplex mPEG-
Fc/CD-PEI/DNA was constructed via host-guest interactions and used as a
control without intracellular dePEGylation. The mPEG-SeSe-PEI/DNA polyplexes
had 4-fold higher transfection fluorescence intensity than that of
PEGylated polyplexes without intracellular dePEGylation. The reason of
enhanced gene expression was investigated by confocal laser scanning
microscopy (CLSM). Due to the cleavage of diselenide bonds, the
intracellular dePEGylation facilitated quick endosomal escape and nuclei
entry, thus resulting in efficient transfection. These results suggested
that the diselenide-linked polycation had promising potential in redox-
