便，包括lac/cat。在药物筛选上，reporter也是广泛应用的。
还有一个是fluorescense。这个在polarization assay和Fret上都可能会有广泛的应用

ABI7500 fast的时候都说什么calibration expried, do you want to continue (
something like this)。
但是相同时间下Taqman assay没有问题。是不是这个机器校正过期对SYBR的影响特别大
，但对Taqman assay没有什么影响呢？
但是确实这个可能是原因，也许需要做SYBR的校正，而taqman assay用其它的filter,
可能还不受影响。 |

底怎么样。但理论上effiency对Absolute Quantification影响很大，而对ddCT方法的
影响有限。而我就只用ddCT方法比较各个gene在不同条件下的表达，没有去比较基因间
的表达。至于ddCt的方法有人要argue有缺陷，不理想，那是另外的话题 (体外话，我
发现没有测primer效率的人不在少数，当然绝大部分人也是用ddCt)。
以一个简单的knockdown为例，设目标gene表达量为X0，在Ct时的PCR产物为Xt0，效率
为EX, reference gene的表达量为R0，在Ct时的PCR产物为Rt0，效率为ER；在
knockdown情况下目标gene表达量为X1，在Ct时的PCR产物为Xt1, reference gene的表
达量不变，仍然是R0，在Ct时的PCR产物为Rt1 (两次的Ct可能一样，也可能不一样)。

除了High Throughput Screening之外，其实还有两种drug design的方法， ligand-
based 和structure-based。前者是在你知道一个靶标有很多结合的小分子时，可以通
过一些软件（例如CoMFA）计算得到一个pharmacophore，然后去搜索具有这个
pharmacophore的其它小分子，再通过实验的screening assay，以期找到活性更好的
leads。
structure-based是指基于靶标的空间三维结构（真实的或homology modeling）进行
rational design，主要包括Virtual Screening，即使用一些软件针对蛋白的binding
pocket （active or allosteric site）进行商业化成百万上千万小分子的docking，
然后通过score和经验挑选排名靠前的candidates，买来做screening assay，筛选到

公司或facility来完成你的HTS,是吗？
如果是，你还希望自己动手参与，你可以找一些大学的HTS center。先看他们有没有合
适你assay的仪器，然后谈价格，以及拿到的hit上你能不能拿出来少量compound作下游
的分析。这个很重要的。一个HTS往往会产生很多hits,你不可能一个一个买回来。大学
的很多center,需要你自己参与，就用center的空间和仪器，center在计算和操作上会
提供一些帮助。
真正HTS还是繁琐的。每天重复，面对几千个hits也搞得人很头疼。你一定要有不同于
第一轮的筛选方法，作为第二轮，这样才能把你的hits变成leads,几十几百个leads才
有可能handle。 |

领域的人用的assay是完全不同的，简直不能互相理解。一般来说，MMS damage repair
更多是用来test BER/NER pathway/factors，研究这些pathway很难用specific
endogenous locus. 对于DSB repair来说,MMS更多只是yeast领域里面用来suggest
epistatic relationship.
从你的描述，感觉你倾向于认为你的locus of interest可能undergo DSB upon
genotoxin stress. 检测最简单的方法就是genomic DNA southern blot for
rearrangements，或者gammaH2AX ChIP，PCR-based assay比如sequencing也可行if

一个是东部的小药厂100人左右，职位是biomarker investigator
一个是西部的仪器试剂公司，400人左右，职位是companion diagnostics assay
development scientist
西部公司的职位职责要求已经定义得非常好了（大公司的缘故吧，跟我之前的工作也类
似），就是测试assay 的一些属性，比如说是不是精准等等。不用做实验，主要是分析
数据和报告。和仪器制造部门交接。不做research，纯development
东部公司的职位职责我其实不是很明白，因为我上一份工作不是做药的。和朋友聊过，
好像不同药厂biomarker做得也不太一样。另外一个就是因为是小公司，可能要做的东
西比较杂，好像也希望我去bench做点实验。

我们使用了文章中报道的高效ssDNA，同时自己设计了其他2条针对体内其他基因位点的
ssDNA，与NgAgo表达质粒共转HEK293T细胞与HeLa细胞，2天及5天后，用T7E1检测切割
效率，这三条针对2个不同基因的ssDNA，在HEK293T和HeLa两个细胞系中，无论转染一
次还是在8到12小时后再转染一次ssDNA，都没有检测到切割。
针对敲除后能使细胞有表型变化的某特定基因，我们设计了ssDNA，与NgAgo表达质粒共
转HeLa细胞5天后，发现细胞表现出了一定的对应表型，但是把这部分细胞收集起来提
取基因组，对靶位点测序，没有发现基因组序列的任何改变。
我们针对某特定基因设计了6种ssDNA，用特异的荧光报告系统检测是否有靶点DSB产生
，结果显示所有实验组均没有检测到对靶位点DNA序列的切割活性。

我们在293T细胞中表达带有Flag标签的NgAgo ，在不同时间点检测NgAgo蛋白表达。发

高端医疗设备，中国还要从欧美进口。什么叫欧美没有基本医疗设备制造能力？
测试剂？你指试剂盒？现在给武肺用的就是Taqman assay (一种real time PCR)，PCR
仪有德国产的有美国产的，你放到中国产，精度能否保证都是问题。精密点的加样枪，
尤其是multi-chanel的，法国德国产，中国产的没法用。耗材，中国产的Eppendorf 管
，pipettte tip，PCR plate都不能保证质量。试剂，就更别说了。即使到最近两年，
中国生物千老用的试剂，凡是要求质量高的，哪怕 培养细胞用的gelatin，还要用国外
的。Taqman assay 用的primer和probe，都是第一天定第二天到，你以为从中国发货。
口罩啥的只不过一时产能上不去。还不是因为全球化把这些低端的产业转移到中国了？
