选antibody或者蛋白需要构建大量的载体提供测试，相应的会出来专门做clone的，专
门做细胞转化的和专门做纯化以及专门做assay的职位。理论上这些职位是不需要phd的
，但根据组员配备和公司要求这些职位中会有一些需要博士学位，一方面博士经验更加
丰富，另一方面博士也是潜在的快速升职对象。既然是大药厂，工资和其它的package
一般和当地大学的AP相当或者略高，这种职位对于在坑了呆久了的博后是一个非常好的
机会，当然也同样意味着激烈的竞争。希望坑里的博后们放低身段，别觉得自己一定要
拿20万以上的年薪，你的实际工作时间工资其实已经比马工高了。
说起找工作，还有一点。千万别眼高手低，你看着职位描述和自己很合，但你真正去面
了悲剧了才会知道自己的竞争对手有多厉害，这个市场上永远有比你合适的应聘者，你
要是能拿到offer只是说明那个时段你合hiring manager的意，没有拿到offer那就继续
加油因为总会有一个喜欢你的hiring manager在等你。 |

this is realy a tough question for those whose major in Molecular Biology.May
be only some guys from Harvard or MIT etc. could answer it,or it is very bad

1. how much power an atomic bomb made of certain amount of materials
could give off.
2. how the power could be controlled.

我们实验室 研究一个 mitochondrial protein的功能。 是一个mitochondrial
transmembrane protein。我的实验目的 是，克隆表达 这个蛋白的 5'-HA tag-Nter
region-Transmembrane-3' (缩写HA-Nter clone) 和 5'-HA tag-transmembrane-C-ter
region-3' (HA-Cter clone) 两个truncated protein.
clone挺简单，因为 克隆cDNA，并且，蛋白很小，只有200氨基酸。我克隆的片段也挺
小的，没有经历太多难度。我把 Nter clone 和 Cter clone 都克隆到 一个
retroviral vector，coding region后面有一个IRES-GFP。plasmid 8.6kb。
接下来,我把这个HA-Nter clone 和HA-Cter clone，用293 cell 表达（CaCl2
transfection）.

条件反复重复。
1 当初选用pQE8也是没有办法，因为pQE9我们实验室没有（pQE8和9的区别就是8在酶切
位点之间只有一个碱基，而9要长得多）。最开始的时候没有意识如此近的酶切位点很
难切下来，最起码同时酶切不可能切下来。但是只尝试过一次顺序酶切不行就没有再尝
试用pQE8了，我不记得是先切BamH1还是Hind3，后面尝试插一段片段在2个酶切位点之
间再切。
Nice alternative, shall solve the problem in this case...but I would just do
blunt ligation, nowadays, the fast ligation kits from different companies
such as Roche, takara, don't really care about whether it is sticky or blunt

Analysis of Genomes and their Expression 的缩写。网址http://
image.llnl.gov. 实际上是一个EST cDNA clones library. 你可以
在invitrogen订购，http://clones.invitrogen.com/
收得倒挺全，但是序列可能会有问题，在3'端常常有错误，出现stop
codon什么的。你这个基因这么大，很难保证有完全正确的cDNA.
当然你也可以选择一些测过全长，甚至可以直接表达的clone库，
同样是http://clones.invitrogen.com. 但是这种库clone十分有限，
你要的基因不一定在里面。另外日本有个免费的克隆库，
http://www.kazusa.or.jp/huge/ , 你可以去找找。
另外，实际序列与网上序列库序列有微小的不一致，其实一点也不
奇怪。我就遇到过这种情况。建议你先问问给你clone的实验室，看
他们的意见 |

读博士的时候，在国内给Bio-rad纯化一种高温DNA polymerase，能解决sequecing反应
时不少二级结构的问题，还挣过几千外快呢。S-35是后来的吧，记得前面用p-32，后来
是P-33，再到S-35。
NEB的catalog确实是一本分子克隆的好书。在国内虽然是作分子生物学出生，克隆没少
作，但来美国作博士后，要作knockout construct时，才真正变成cloning expert：
pUC18里要塞进一个20kb的片断，找合适的restriction enzyme都困难呀。记得当时就
一个Not I是识别八个bp的酶，很有用，另一个是Sal I。Sal I是识别六个bp,但在老鼠
genome里，不知什么原因，位点很少，几个constructs的最后一步，三片断连接都是两
边用Sal I和中间另一个酶。有一个cloning strategy, 整整两天都没有想出来，只好

如果你下一次做类似的课题，我建议你用Gateway cloning system，只需要将你的基因
克隆到pENTR vector（RE cloning，LIC cloning, TOPO cloning or BR
recombination），然后你就可以随意的将你的基因克隆到其他带有不同tag的pDEST
vectors上了，效率很高。
其他的还有ligation independent cloning (T4 DNAPol)和In-fusion cloning。 |

would think it over and over like with any complex experimentations: what
are the options and what might be a best one to approach this...As far as I
am thinking, the less passaging the better...otherwise one might be running
a risk of picking the cells from a same clone multiple times; as well, the
clonal growth/death bias over the passaging could also dramatically change

第一篇 DNA测序， Sanger Style - 纪念Frederick Sanger
2013年11月20号，Frederick Sanger 去世了， 享年95岁。Sanger是DNA测序之父，获
得过两次诺贝尔奖， 一个是1958年蛋白测序， 另一个是1980年DNA测序。虽然DNA结构
发现的比蛋白晚， 但毕竟是genetic code，master script。ATCG四个碱基的组合比蛋
白的20个氨基酸的天文数字组合可更容易研究找规律了。人天生就是个好奇的动物，
最喜欢找规律, 尤其喜欢从简单的地方入手，所以围绕DNA的研究在Watson-Crick的碱
基配对这个简单地规律被发现以后，起来的就比蛋白快了很多。其实DNA和蛋白， 就像
