我从种子提nuclear extract, 想做EMSA, 用的是DIG-Gel shift kit. 开啥有了
TATABOX （5 repeats) 25bp in length, 试试看蛋白是不是有功能的，结果没加蛋白
的free probe也有带，所以认为我那加了蛋白后产生的带不是真的binding，因为2 带
的位置是一样的。但是有些情况用其他大一点的探针，50bp, 会得到smear. 还有我跑
电泳的胶很小，8x8cm（bio-rad)， 5% native gel. 0.25x TBE buffer, 4C, 80V.
问题：
1）怎么能证明蛋白是用功能的？现在没有binding， 不知是不是蛋白的原因，SDS-
PAGE 检测蛋白还行。抽的几次都是结果差不多，尽管有些带很强且集中在2个区段，但
总体来讲，蛋白的分布大小都有。
2)能不能找个其他probe 做postitive control。 有人可以推见一下啥探针？
3）我是用的nuclear extract, 探针可不可以用200bp 长的。
4)调整些啥才能有好 |

，potency比较高些，另外液态的比固态的potency高。grapefruit对肝肾detox很好，
可惜国内很难见到grapefruit或grapefruit汁，所以你可以带些grapefruit seed
extract回去。

液体的，小瓶子装的，一般会在买液体调料那里。缺了这个香味会少，但是不知道会少
多少。
我觉得是不是应该加些butter/melted 替代食物油，糖加的太少了，要加量，至少要多
加1/4cup of sugar，而且用brown sugar颜色和味道都会好。
你的baking soda是不是放的table spoon呢？ 如果是吃饭的勺子，那绝对是太多啦~
还有就是做这个简易面包或者banana muffin一定不要用搅拌器，是大忌，就把啊材料
都混到一起就好，如果搅拌了那么就会很dry，dense
还有一些就是我自己的小建议，可以加一些sour cream，就是酸奶油买cheese的地方都
能看到，然后可以用lemon zest（lemon皮用那个孢子擦点皮屑下来，代替vanilla
extract或者互相加强） |

泡芙很简单，里面的绿茶奶油馅及其好吃，不甜不腻~

一下分量我一共做了20多个，大烤盘分了2次烤，我家烤箱一次只能烤一盘>.<
cake flour 100g，water 160g，unsalted butter 80g，sugar 1tsp，salt 1/2tsp，
egg 3(实际上大概用了3个半）
425F 12min, 待泡芙膨胀定型后，将温度降到350F，25min，直到泡芙表面黄褐色。
制作过程：
1、水、盐、糖、黄油一起放入锅里，用中火加热并稍稍搅拌，使油脂分布均匀。当煮
至沸腾的时候，转小火，一次性倒入全部面粉。
2、四根筷子握在一起来搅拌，使面粉和水完全混合在一起，一定快速的搅拌。
3、一直搅拌到面粉全部和水分融合在一起，不粘锅以后，再关火，可以把锅从炉子上
取下来了。（此时面粉全部被烫熟了）

参考分量：6寸蛋糕盘一个
配料：一袋lady finger，bailey's 咖啡酒200ml,黑巧克力70克，unsalted butter
125克，细砂糖70克，vanilla extract 1/2小勺(2.5ML)，almond meal 125克，cool
whip 150克，草莓或者raspberry或者罐头黄桃适量
配料中，酒的含量可以降至150ml左右
黑巧克力一定要用好的，因为这个蛋糕完全不用gelatine，只是用巧克力来凝结，一般
的chocolate chip凝结力不够强，又太甜，我用的是trader joe's买的pound plus
dark chocolate
水果的话，用草莓或者raspberry应该最为合适，罐头黄桃有点甜，跟巧克力味道不够
和谐，但是我小农，最近草莓raspberry贵。。。

发帖数: 720 | 44 可能是因为冬天的来临，我越来越爱巧克力的甜点。souffle的杯子买了很久，烤过布
丁，烤过软心蛋糕，就是没有烤过souffle，原因就是我总觉得 souffle的方子太难，
有太多需要注意的地方。如果不是这次为了处理多出来的蛋白，可能我还是不会做
souffle……
翻了好几个方子，最后决定用巧克力而非可可粉来调味
配料：
蛋白2个，蛋黄1个，sugar 3tbsp，dark chocolate 55g, heavy cream 2tbsp，cake
flour 25g，vanilla extract 1tsp, bailey's 1tsp
过程：
1、蛋白加糖打至干性发泡
2、heavy cream 微波炉加热15s，倒入巧克力，搅拌至完全熔化
3、巧克力奶油稍冷却后倒入蛋黄，vanilla extract, bailey's 搅拌均匀
4、面粉过筛，用橡皮刮刀拌匀
5、像做戚风蛋糕那样把蛋白与巧克力糊混合均匀
6、souffle cup杯壁用黄油抹一遍，全部都要抹到，然后倒入5的混合物
7、把烤杯放入装了开水的烤盘，350F, 18分钟

香草冰淇淋:
1 cup whole milk
3/4 cup 糖
2 cup heavy cream
1-2 teaspoon pure vanilla extract
先把糖加到牛奶里, 搅拌到糖溶解. 然后加heavey cream和vanilla extract, 搅拌好
之后放到冰淇淋机里, 20-25min
我按这个配方做过香草的和草莓的, 冻起来之后看上去硬硬的, 实际上很creamy.

holiday dessert，cranberry bliss bar， 感觉很不错，就在网上google下，据说这

有翻糖皮的缘故也不易损坏，所以制作好的翻糖蛋糕室温保存2-3天是没有问题的，一
般也不建议冰箱存放。
翻糖蛋糕的主要装饰材料自然是翻糖（Fondant），它的全称其实是Rolled fondant，
对应的还有一种叫Poured fondant（一种不太常见的甜点夹馅）。因为前者在蛋糕装饰
上非常常见，现在往往简单地用Fondant来代替了。市售的成品翻糖，一般是用食用甘
油（Glycerin）加糖粉（Powdered）和吉利丁（Gelatin）还有少量其他原料所制成的
，因为它的特殊质地（像橡皮泥）便于造型，出现之后迅速流行，成为婚礼等各种重要
的庆典场合所用蛋糕的装饰主角。它的出现，让“只有你想不到的，没有你做不出来的
”造型成为可能。
我这次购买了两个品牌的翻糖，差别很大，各有特点。覆盖整个蛋糕表面的是
Fondarific牌子的香草味古典白色（classic white）翻糖，2磅重。是象牙白那种带一
点点微黄的白色，其实这种卡通造型的蛋糕如果用纯白效果会更好。它的质地非常柔软

，节食一个星期，，原本打算今天早上去健身课再运动一个半小时，我就可以心安理得

上净是她的动态，果不其然，昨天出了通知：今日的健身课提前一小时！NND，我满怀
一腔悲愤回到家开始做这个蛋糕，胖死我算了，星期一再减肥！
配方用的是Cook's Country 系列的Blue Ribbon Desserts，上面那张成品照片的背景
