反而是被切过的(或者alternatively splice) 的可以被pulldown.
如果你开始用的是native buffer pulldown 的话,不妨换成denaturing pulldown
condition
比方说用 2 %SDS 把细胞给煮了,然后加缓冲液稀释到0.2% SDS再pulldown,
这样一方面可以解决大部分构象原因,另外一方面也会减少背景
当然,如果你比较倒霉的话,这样也有可能导致你什么都pulldown 不下来。这个你得
试一试,就没有人敢给你保证了。 |

pulldown result by WB? In such case, it would look decent.
In the second case were you trying to pulldown in vivo synthesized protein
with isotope? Indeed, the signal/noise will look awful, because many
endogenous protein also have the radioactive label.
Well, if that's what you want, you probably will need to try the anti-6XHis
antibody. Unfortunately I don't know which antibody works for IP. You should

听说把beads先和antibody cross link一下,可以提高pulldown efficiency;

version of your protein, is that right? I am concerning about that your
GST-fusions are so geographically close to each other there will be very
little chance to form a GST-protein/protein dimer. Most of them will be
GST-protein/GST-protein dimer, that's why "E coli. 表达纯化的yeast蛋白不能形成
二聚体 (GST pulldown)".
But I'm just speculating, didn't know your exp. detail.

without sucrose) following by gel filtration to remove free labeled AAs.
the problem is the pulldown assay signal is very low. I may try both NTA-Ni

If you have a doubt about this, you should load the eluant directly without
any pulldown to see how much full length protein you recovered.
I don't know why you are bothered by the free amino acid. They will be
washed out in the pulldown process. I suggest you NOT to do the gel
filtration at all.
What you need to do is this:
1.centrifuge to pellet the ribosome. Are you sure you are using 100,000 rpm?

page gel上都有信号。 但是pulldown后没有信号。主要基于 Farnham protocol。
他的ChIP buffer就是RIPA,beads也没有block,这个没问题么?有些别的protocol用
salmon DNA block beads,salmon DNA的methylation会影响 DNA methylation dot

After you login to your Kohl's online account, move mouse over "ACCOUNT"
under your name on top central of webpage. From the pulldown menu, click "
Yes2You Rewards". Then you can sign up. |

菜鸟健身入门!
3年前刚来美国的时候,根本也没想过要健身什么的,刚开始学习也很忙,自己做饭做
得太好吃,结果一年多长了40磅肉。暑假去海边玩,拍的照片不能看了,呵呵,当时也
没想那么多,后来发生了一些事,打击很大,心情比较消极,需要转移下注意力,就开
始锻炼了,一练就是一年多,分享点自己的经验,入门级别的。要修炼神功,自宫倒不
必,有些装备还是必须的。
装备:
手套一付
助力带(strap)
Lifting belt (护腰) 有的健身房有借,可以省钱哈
战友:
一个人容易偷懒,有个人或者几个人一起,练起来会好很多。
药品:
刚开始没使用,到了集中减脂的时候采用。
蛋白粉(吃了但是不知道有没有用)
肌酸(还是有点用的,能提供能量,减脂的时候,吃得东西少,肌酸能补充能量,使肌
肉受到比平时大的刺激)
NO (这玩意没用过)

在gym称还是109lb,在游泳馆称又回到107lb了,希望是gym的称不对,呵呵 |

跑步机20分钟,4.0+6.0mph
elliptical 10分钟, level 8
力量几乎没做,就一个,好像叫lat pulldown?
然后就是拉伸,垫子上做了一组pilates的练习butt的动作 ....

然后吃了一份香茅鸡饭,真是yummy~~~ |

先是一个小时篮球,期间被一个黑人大壮称赞说 This guy knows how to use his
shoulder,另一个黑人高佬看到我抢篮板的积极程度竟然对我高呼今年NBA选秀去芝加哥
的佛大大前锋的名字--Noah。虽然我知道他们有过誉之嫌,不过还是着实为自己的力量
进步得意了一把,哈哈。
之后是腹部训练,135个CRUNCH; 最后是跑步,以放松的8麦速度跑了30分钟
星期六:
引体向上三组:10,8,6个
PULLDOWN三组:115*12,120*10,115*10
T-BAR:45*12,55*12(第二次做,重量还控制不好,感觉还是轻了)
直腿LEG PRESS三组:290*15,310*15,350*15
本来还想接着做硬拉和LEG CURL的,可惜关门时间到了,只得依依不舍的痛别GYM。星
期天一定要恶补大腿后侧的力量练习 |
