我再用g-75resin去分离蛋白的时候遇到很多困难。有时候resin用一次后速度变得非常
慢，低于0.3 ml/min，有时候back pressure很大，以至于柱子都爆裂了。请问用g-
75resin分离蛋白的时候要注意哪些地方避免堵塞。用过得resin怎么保存？谢谢！ |

30min). But, normally those commercial NHS ester has a spacer between the
resin surface and NHS, which allows further functionalization of the resin
with proteins. Anyway, any info. about 5~10 um -COOH resin is also

-scale的时候用的是NEB的streptavidin magnetic bead，效果非常好，而且很方便。
但是现在做large-scale, 感觉太贵了。之后试了pierce的streptavidin cartridge 和
resin, 都没有NEB的分得干净，而且还有20-30%的损失。我不确定是我操作不得法，还
是pierce确实没有NEB好用。
烦请前辈给推荐个好用的产品或者指点一下方法。

都要拖。准备自己填一根大玻璃柱，看了一下resin，只查到两家：
BIO-RAD的polystyrene beads和Tosoh的toyopearl的hydroxylated methacrylic beads
前者理论上来说是完全hydrophobic的，后者表面有OH，理论上会有吸附？
就我的用途来说是不是应该用polystyrene的比较好？
还有什么推荐的vender吗？BIO-RAD beads的operating range跟我的体系不是特别地
match

要做deep cleaning和补9个牙，不过医生要先deep了再补。。。好像严重得就三个左右

医生说要给我加半个小时因为牙垢多，多收25快 （医生说保险不cover这个extra），
所以一个quad是253，一次下来是1000多，自己可能要给400。

（十三）装柱子的学问。
上篇提到了Gel filtration的基本原理，现在谈谈我装柱子的过程。我们用的是Amersham
的 XK26/40的空柱子，然后往里面填充Sephacryl S-300 HR 的resin。按道理说，要提高分辨
率，柱子应该是越长越细才好，这种柱子虽然不短，但是挺粗的。但是一个好处就是，样品体
积可以大一些。
装柱子本身就是一件很tricky的事情，以至与这课的教程上专门列了一小节来解释怎么装
柱子。我们这一组人中，我负责装柱子。一开始就是洗resin了，用粗制烧结玻板过滤的漏斗来
洗，感觉这方法还挺快的。洗完了，就把resin重悬成50%的slurry，然后就是往柱子里头倒了。
但是呢，有一个小窍门，就是必须先封闭柱子的下出口，往柱子里面倒10%体积的buffer。我
过去装柱子犯傻，不知道要这么做，结果发现下面的resin 不均匀不说，还有一大堆气泡。所以
先加buffer是很必要的。倒slurry的时候还得用玻棒引流，这样可以尽可能的避免气泡。然后就
是等了，等resin 沉降３０分钟后，就可以打开柱子的下出液口，让多余 |

2：它是复杂的混合物，它的物理性质与原油的性质有关。其主要成分包括Asphaltenes

3：工业用沥青的生产过程（air-blow），也会对其物理性质产生影响。
未 blow 的沥青softening point 大概是54度C。，3rd blow可达到174度C. 随着温度
升高 Asphaltenes含量上升，hard resin, soft resin, oil, waxes含量下降。 从而
使得沥青越来越硬
4: 沥青是个统称：有很多不同的产品用于不同步地，铺路，铺屋顶。。。。由于生产
工艺不同，因此，物理性质也不同

发帖数: 2998 | 38 之前有一群人对比过各种Web Server的速度。好像JDK 1.5之后的Tomcat的确比很多纯C
写的Web Server要速度快(除了resin外)。
然后通过仔细分析，发现这些纯C写的web server主要的瓶颈在于内存管理。比如每次
新请求进来之后，总是要malloc,然后处理完之后再free。而JDK1.5之后在实现对象分
配的时候，只要几条机器指令就完成，速度远比C/C++的内存块链表遍历快很多。然后
JVM实现的统一内存回收，一次性全局回收比离散多次free要高效。
但是唯独比不过最快的resin。因为resin的作者自己实现了很高效的内存池管理方式，
它可以在程序启动之后，到退出前不做任何一次malloc和free。
所以，很多Java号称比C/C++更高效，主要就是在于内存管理这块。能够做到像resin的
作者那样精湛的C/C++功底的人毕竟很少。所以，一般设计到多次内存分配和回收的程
序，还是尽量用Java比较方便。

发帖数: 28562 | 39 这个很难做，属于我最讨厌的实验之一。最大的误差来源于洗resin的时候损失。
我一般是这么做的：
1. 把放抗体前的sample 调到同样体积， 然后取等量小样留下来分析 (S-0)
2.放抗体，blah blah blah 按照经典方法作。
3.放过量的resin。 可能的话混进等体积的blank sepharose bead 来增加体积。
4.把所有东西都装进mini-column 里面，等柱子成型后收集flow through.
然后把原来的管子用buffer 涮涮之后全部加到柱子上去以保证搜集到所有resin.
5. 洗柱子
6. Eluate：用1 volume 2XSDS-buffer (95oC) 洗脱，重复三次。
然后把柱子装到５０ml conical tube, spin down for 1 minutes, to collect all
the solution stuck in the resin bed. |
