strand specific:
1. strand-specific or not 不影响mapping，因为不管用什么protocol，出来的reads
map到哪条strand都算。再算gene expression的时候才会用到strand information，
具体怎么count reads to gene, 这个要取决于你的strand specific library prep
protocol
2. 拿tophat/bowtie来说，mapping的时候有一个选项--library-type，这个怎么选都
不影响mapping，但是一定要注意，接下来用cuffilink/cuffdiff和htseq-count之类的
，这个--library-type一定要添对了，这个影响gene expression的counting

时报》文绉绉地说他的诗 “imparted an unsettling sense of personal
dislocation”，就是评论家Richard Howard说的“the working of the divided self
”——那其实是说这位诗人精分错乱啦……
他在1964年出版的第一本诗集叫《Sleeping With One Eye Open》，够瘆。其中有他最

上周末去NYC一趟,到了Union Square,想起版主多次提到的Strand Bookstore不就在附
近吗？向一个晒太阳的大叔一打听，沿Broadway走两条街就是.
这里是Broadway和E12街角.后面绿树葱葱的就是Union Square了.再远可以看到帝国大
厦．
书店外屋檐下有不少特价书.书中就有颜如玉.先给mm拍一张.
摄影书在二层的楼角,光线很好,窗口有四把椅子，可以坐定慢慢翻书.
（椅子边有插头，赶紧把用了一半的相机电池插上了.呵呵）
书满满好几架,按摄影师分类.书的种类太多了,
听说过的有,没听说过的更多，几天几夜也看不完.
而且一本书是新的和二手的摆在一起,买不起新的可以买二手的.
新书一般都是一本随便翻,其余在塑料薄膜里包着.
窗户都是打开的,有新鲜的风吹进来．这里窗台上有艾未未的一本,

illumina的kit是在second strand cDNA中引入dUTP,然后amplify library的时候这条
链不扩增.
NEBnext的protocol是用USer 酶把这条链degrade,然后再扩增
这俩有什么区别??
我准备用KAPA的kit,因为这个kit不带adapter,需要自己买adaptor,本来想订NEB的
adapter的,但是NEB的protocol又不一样...彻底糊涂了,哪位大侠指教下?不胜感激! |

我的初半马是八月份，成绩是2小时30分钟。我当时定下的这次半马的目标是2小时15分
钟。
比赛前一天，我查看我的训练记录，发现自己的Long run 都在每一迈12分钟左右的
Pace；有几次Tempo run 倒是10分左右的Pace，但是只能跑6迈以下。我心里直打鼓，
这个2：15的目标是不是不太靠谱呢？但是回想起在跑版看过的几个出色的比赛报告b大
家都在关键时刻激励自己挑战自己，我心中闪出“明知山有虎，偏向虎山行”的大字，
于是做好一个2：15的时间表放在手腕上，就睡觉了。

5. 在哪个权威网站或database比较容易找到一些实验的原理精解？
