发帖数: 1187 | 2 土人第一次做，全盘follow trizol的protocol。为省reagent用了100mg tissue
per ml trizol。发现以下问题，请达人指点：
1. chloroform extraction后的precipitation，为什么用isopropanol？室温
incubate也让我很困惑。。。
2. 做了两次，第一次是heart，拿到RNA很顺利，测OD时发现：260 peak偏向270；
230附近有极高的峰。看来chloroform和precipitate后wash的步骤都不太effective？
3. 第二次做liver，搞到巨多RNA，很oily，dry了之后pellet死活不溶，试了
60oC加热，加大volume，最后还是有很多不溶。是不是trizol用少了？
4. 用了hand held homogenizer和pestle+mortar两种tissue homogenization
methods，run了urea agarose gel（顺便请问大家都用什么denaturing gel），
18S:28S rRNA大约1：1，很困惑degra |

or more specifically IgGs out of the tissue so there will be much less
background in IHC.
Correct me if I am wrong
For embryos, endogenous IgGs are relatively low compared to adult tissues.
Maybe that's why it works well without perfusion. |

发帖数: 1841 | 15 一个knock in 的老鼠，目的基因与GFP融合表达。我的目的是让GFP和其它抗体共染色。
发现如下：
Harvest tissue，fix with 4% PFA overnight at 4 degree，切片后GFP无需染色可见
微弱信号，但是其它的抗体无法染色
如果直接用OCT在干冰上速冻，切片后再用4% PFA固定，其它抗体染色没有问题，GFP的
直接信号不可见，GFP抗体染色后一片smear，但是与control无GFP的tissue相比可明显
看到smear的GFP信号。
如果fix with 4% PFA for 2 hours at RT，然后incubate with 30% sucrose
overnight，那么GFP和其它抗体的染色都没有问题。
虽然我根据别人的protocol找到了解决的办法，但是一直想不清楚为什么。
为什么4% PFA固定过夜其它抗体就不能染色？为什么速冻后再用4% PFA固定，GFP信号
减弱或者变得smear？为什么固定两小时再用sucrose处理，则GFP和其它抗体的染色都
得到恢复？
大牛出来解答一下原理。 |

在我们这里，一个Tissue Engineering的教授以前就是化工系的，
(他自己当年拿的就是某牛校化工的PhD学位)；
10年前学校组建bioe的时候，他是当时bioe的核心力量之一，
所以他既属于化工，也属于bioe
相应地Tissue eng既能算bioe,也可以算化工吧 |

单的问题：用PLGA为材料，要你做一个scaffold，必须满足时间(growth factor
controlled release)，空间(内部和外部)，组成,机械强度可以控制。还不能加热，不能
用有机溶剂。你现在能做的到吗？我现在可以说我们基本已经完成了这些指标。但我们从
不发文章,我认为发文章是很无聊的事情。你以为Harvard MedScool的文章惨不忍睹。人
家没有把好东西拿出来。首先你从文章来看一个行业的发展，就完全错误。发文
章都是要有经费(当然，混博士的除外)，而每个经费都是要有一个项目，而每个项目都是
有一个大背景的。我来给你分析一下。Tissue engineering的终极目的就是替换器官。那
好，人体都有什么器官？首先是皮肤。人工皮肤也是目前为止唯一成功的Tissue
engineering产品.再就是心脏，当时有个project叫“LIFE”，聚集了一大帮人，最后失
败了。MIT的人工肝脏，最后也失败了。
