liver里提取RNA我也出现相同的状况，非常难溶，一般我会根据pellet大小加多点DEPC
水，如果还是不溶，我也不管它，貌似放到-80度储存后它最终会自己溶掉的。要么我
就多vortex一下。
有个数据说1mg的tissue中，muscle或brain等大约可以得到1-1.5 ug的RNA，而liver可
以得到6-10 ug的RNA。所以巨多RNA正常。 |

直接做PCR，PCR run一个半小时，跑胶20min。
今天听说Ambion也有一个RNAlater solution，把tissue放在里面很稳定，据说很牛。
我们就是想看看mutant里面哪些基因的RNA水平有变化，然后跟phenotype联系起来好找

啊。
做library的话facility用的是illumina mRNA sequencing sample preparation kit，
两天完成library，似乎不用花太大力气。有没有做过的朋友？ |

reporter mice. Very nice GFP fluorescence signal was seen in ductal trees

digestion). No GFP-positive cells were detected using FACS analysis. Can

reagent也提取到了mRNA,nanodrop测出的浓度有800ng/ul，但是我试了cyclophilin，
gapdh，18s几个内参，都测不到CT值。已经重复几次了，郁闷的不行。请教各位，有什
么建议么？
SVF protocol
1）isolate WAT,
2) Incubate tissue @ 37C for 45min with shaking @ 80 rpm （Collagenase I，
1mg/ml）
3）300g for 10min
4) Dissolve pellet in Tri-reagent. |
