Recently I am doing drug uptaking experiments. I put the swollen hydrogels
into the drug solution. At different time intervals I take out some drug
solution, and dilute them to detect the concentration by HPLC. First I thought
this way is good to determine the drug uptaking kinetics. However the results
are not good because the concentration of the solution didn't change very
much.
Can you tell me how to determine the drug uptaking?

Do you think both water and drug diffusing rates are same?
If not, water is faster, the concentration of surrounding solution will
increase. How can you get the uptaking kinetics by determing the concentration
of surrounding solution?
What kind of method I can use to determine amount of drug uptaking except to
release?

What is the weight ratio of the drug solution to the swollen hydrogel? If this
ratio is too high, you should expect that drug drop in the drug solution
would be very small. How about use less drug solution but high drug
concentration? BTW, why do you want to do this kind of drug uptaking? at first
, I though it's about drug uptaking by cells!

I want to check experienced parents how you feed your toddlers (2-3 years
old) with sufficient amount of milk every day? I usually feed him milk 3
times a day, morning, afternoon and evening before going to bed.
It is fine until now he is potty training. I am trying to reduce the
urinating habit during night sleep, so don't want to offer him the milk
before going to bed. Then it is difficult to maintain the same amount of
milk uptake.
Want to check other parents how you solve this? Thanks.

上来问问各位生物同学,实在没有办法了。最近试验发现小鼠皮下注射后一些lipids能
很好的留在淋巴结里,主要是macrophage的uptake,初步试验似乎是albumin和这些
lipids高度亲和,将它们带至淋巴结,但是albumin是如何转运这些lipids到
macrophage的呢?如果是nonspecific cell membrane insertion, 似乎淋巴结里其他
的细胞(B,T,Cells)因该也会分一杯羹。我的问题是:一般说来,lipids或者是
fatty acids是如何被lymph node macrophage 捕获的?有没有cell-surface receptor
在macrophage细胞表面?多谢,我不是学生物的。

是有少部分DC in lymph nodes 也uptake,但大部分都是macrophage,皮下注射2小时
后淋巴结里就有大量的lipids,所以主要可能还是lymph node draining而不是兰格汉
DC transfer.

巨噬细胞应该是肯定可以uptake的,不起知道肝脏和脾脏。有经验的同学指教一下吧。

你说测血液、肝肾里面的细菌含量,意思是可以检测到活的还是死的细菌?还是细菌被
降解的代谢产物?
另外一个问题:细菌被巨噬细胞uptake以后会降解,这个降解之后是不是连细菌的DNA
都没有了?

终于得闲可以坐下来写点东西了,喘口气先。
首先,这篇评论是目前我看到的写得相对最中肯的一片,原因很简单,作者确实是这个
领域的人,且就事论事,没有想很多人那样屁股决定脑袋。文章的开头和结尾已经给作
者的态度清清楚楚定了基调。所以有人希望用这篇评论来扯什么“皇帝的新衣漏洞百出
”的淡,请回头再仔细看看作者自己的是怎么明确表态的。当然某些人惯于断章取义,
这些人不早生个40-50年真是太浪费了。
闲话不表了。一篇好paper,并非一定是十足完美而没有质疑的,但一篇好paper一定是
经得起质疑经得起时间考验的。眼下很多实验室包括作者包括我自己都正在基于NTCP这
一新发现进行新的研究,时间自然会说明一切。
这篇文章,指出了原paper中的诸多不足,其中的一部分我也是很同意的,事实上拿出
任何一片CNS paper,基本都可以照猫画虎的说出个123点不足和缺陷来。至少我们在开
journal club的时候,无论出自哪个杂志/谁人之手,还没有哪篇paper能幸免的。重要
的问题是,这些质疑和不足,是否根本性的动摇了文章的基本结论,还是只不过是需要

(1)卫生部2007年颁布实施《新资源食品管理办法》有意取消了2002年颁布实施
的《转基因食品卫生管理办法》“第五条 转基因食品的食用安全性和营养质量不得低
于对应的原有食品”这个条款,是退步,而不是进步!这种删除的实质是,进一步顺从
邪恶孟山都为首跨国转基因产业的私利与压力,置中国人民持续安全、健康、生存与繁
衍于不顾!
(2)卫生部2007年颁布实施《新资源食品管理办法》有意取消了2002年颁布实施
的《转基因食品卫生管理办法》“第五条”,但是卫生部《新资源食品管理办法》“第
三条”规定:
第三条 新资源食品应当符合《食品卫生法》及有关法规、规章、标准的规定,对
人体不得产生任何急性、亚急性、慢性或其他潜在性健康危害。
依据该第三条规定,在对于抗草甘膦转基因大豆加工的转基因大豆油,检测出超过
俄罗斯对大豆油草甘膦残留量允许标准与中国卫生部对棉籽油草甘膦残留最高限量2.9-
6倍的草甘膦残留量,已经证实对人类健康,特别对青少年、儿童、婴儿与孕妇及其胎

宋的文章够了,experience不够让人信服。
宋的博士学位在中科院拿的,在us做了3-4 年postdoc。宋在us 待的时间太短,对美国
博士培养体制和科研体制先进之处了解得可能不够。一同面试院长的有几个candidate
文章比宋好。武大可能是看到宋在中科院当个官,搞个管理就把it请过来了。武大应该
找一个在美国拿个PhD,然后在us 做postdoc后海归的当院长。
宋自己的科研做得很好,但对宋领导一个院做出像样的改革,大幅提高整个院科研水平
不太看好。

I've checked that paper. Based on my understanding, the paper said ATP
stimulation can temporarily inhibit respiration and reduce mitochondrial
membrane potential, so inhibit mitochondrial Ca2+ uptake. After temporary
inhibition of Ca2+ uptake, ATP is consumed and mitochondrial respiration
resumed and cause mitochondrial Ca2+ boost. And the boost after ATP
stimulation can reflect the ability of cellular respiration.

genome里面,还是细菌uptake人的食物里面的某些DNA片段?
你前面说cooking没有完全破坏DNA。。。完整基因基本没有了,但是没错基因片段是有
的。但是你知道这些片段都是不functional的。你做这么多knockout这是常识吧。然后
你推理出因此人可能会把大米转的基因加入人细胞。俺们告诉你不可能。
然后你接着反驳说病毒会加入人体。没错啊,可是大米怎么改良转基因也不是病毒。
然后你说细菌跟细菌间可以转基因。也没错啊,可是大米有不跟细菌转基因,人也不跟
细菌大米转基因。跟吃大米有什么关系?

1. RAI后做的是whole body scan吧,你确定是PET?
2. 有的aggressive的肿瘤对I131不敏感,所以没有uptake
3. TgAb如果baseline就高,不说明问题,跟TG高是两回事情。
4. 淋巴结原来做手术前有吗?术前应该有详细的颈部超声,影响外科手术清扫淋巴的
范围。现在需要穿刺确诊。
5. 甲状腺癌的治疗手段很limited,如果I131不敏感,有复发,可以考虑参加clinical
trial, 现在有几个药可以试试看的。
6.甲状腺癌带瘤生存的情况很多,有时候就是没有办法根治的。
