orthosteric agonist的dose-response curve, 完全相同的浓度梯度和条件. 但是3条
彩色的curve是叠加了compound的curve, 不同颜色表示不同浓度, 但是在同一个curve
下,compound的浓度是一样的. 回头查了一下以前的数据,似乎在这些浓度下,compound
自己有弱效果. 所以这个结果有可能是效果叠加的结果. 没法说明到底是不是
allosteric modulation. 是这样吧.
撇开这个图. 从头想想. 我有个compound, 与receptor的 binding affinity 是几十个
nM级. radiolabel的competition assay, 看上去几乎80-90% replace orthosteric
agonist. 问题是,测了compound自己所有的下游信号, 几乎都是negative,或者是uM级

inhibitor，像一些kinase的inhibitor都是ATP的类似物；即Mechanism-based
inhibition，共价和非共价结合的都有，academic groups 开发比较多。但这类
compounds选择性可能不好，对同类型的酶都或多或少有抑制作用。
另外，从已经报道的ligand去设计similar analogs，可能很难跳出别人compound的骨
架(scaffold)，发文章的novelty或专利申请受影响。
如果是靶向protein-protein interaction 的话，尤其是你知道它们的complex
structure，以其中一个为receptor，另一个的peptide为ligand，不断优化（突变和/
或改变长度）这个peptide，使其达到活性达到最优；并且可以再对这个peptide进行化
学modification，提高其透膜性或水溶性。

开辟了一个新的应用方向，所以有一些后续的工作，还有一些是在实验上的应用，也有
一些review和书收录，不过大段介绍的不多。
作者背景get，，另外接着应用的问一下，除了被1. 新合成引用，2. 实验应用外，还
有哪些是可以证明compound影响力的？我只能想到被工业界生产，那么化学合成的一些
新compound如果被工业界生产出来这个事情怎么查？compound到进入市场是怎样的流程
?
以上是significance, 就是影响力。另外在归纳originality & contribution时这种工
作要怎么总结呢？ 就是说自己合成了compound 1，2,3，、、、吗？这样一句话就可以
说完了，要怎么展开说这样的contribution？让IO看起来感觉份量重一点？

我对于药物销售的保护还有些不明白的地方，请大侠指教。
我的理解是保护有两个，一个是专利，特别是compound本身的专利，大概20年吧，还有
一个是product的market exclusivity, 如果是头一次批的就是5年，orphan drug 好像
是7年。那么这两个是叠加的呢，还是哪个长算哪个？比如说，
对于一些老化合物来说，compound本身的专利早就过期了，这种情况我想就是靠market
exclusivity来保护药物销售。如果是个很新的compound, 本身专利20年，整个开发过
程可能10年，批准的那天起，专利还剩下10年，也就是说，market exclusivity那5年
失效的时候，compound本身的专利还有5年，要等到批准上市10年之后，也就是
compound本身专利全部过期的那天，generic才可以开始销售，这种情况innovator药物
销售是靠化合物专利来保护而不是market exclusivity
不知道我的理解对吗？请方家指正，谢谢。 |

如果取expected annual return == 14.10%
SD on annual return == 16.70%.
Years == 30年。
做了100000次随机模拟。结果见图（值是目测的，不一定精确）：
expected compound annual return == 12.85%
Standard deviation on compound annual return == 3.08%
图形不完全像是一个正态分布。
Ringer的表格里的Standard deviation on compound annual return得以验证，
expected compound annual return 不知为何模拟出来的值小于14.1%。 |

APR = (monthly rate + 1) ^ 12 - 1, I guess you refer annualized compound
interest rate.
In some sense, you are absolutely correct. However, normally, but not always
, in the mortgage industry, (effective) APR is the fee + compound interest
rate.
In order to let borrowers shop around, APR is used to have the same benchmark regardless how the combination of cost and rate is evolved. ($2000 3.875% vs. $5000 % 3.125%)

补洞。我找了个plumber来检查过确定没有leaking之后，他用joint compound把洞给补
上了。但问题是，补的那部分很明显的凸出来，和周围不平，而且颜色和纹路也完全不
协调。Plumber要让我加钱，他需要再来3大桶的compound把整个天花板刷平。

They are titled as one single family house.

different method, such as RT-PCR, or WB? Normally, people will assume a
compound with no dose response as a false positive. But it might also be
possible the compound is so potent that EC50 is lower than the lowest
concentration you tested. You might need to tell us more to let us help you
guess.

乎不太实在吧!听上去象GENENTECH在倒买倒卖你家的COMPOUND,但这两家是有合作协议
的,如果有人要你们的COMPOUND都是要按照协议上进行MATERIAL TRANSFER.另外
GENENTECH的绝大多数MTA都是提供免费的COMPOUND,而不是拿去卖. |

curves是比较现代的用来发表的东西，你为什么要转化？
就这个图来说，B很可能是negative allosteric modulator，因为competition curve
达到的plateau跟你用的agonist有关，说明B不是competitive binding。如果B是
competitive binding，理论上当B达到足够的浓度应该会完全compete掉agonists从而
最后的plateau完全是B的效果，如同A一样，agonist-independent，现在B的
competition curves plateau是agonist-dependent，说明B是non-competitive
binding。
当然，证明一个compound是allosteric ligand是复杂的，一个实验不够，往往需要多
个实验去从各个方面证明。就你贴的这个结果，很多问题，首先agonists应该不是

of the cargo carried by the transporter? You don't have to do this
experiments in lysis buffer. The transporter protein, ideally with a tag can
be prepared from oocyte, then you throw the labelled compound in the oocyte
system. After pulling-down the transporter, the binding can be analyzed
through assaying the amount of compound incorporated into this transporter.

筛选系统，筛选出来的compound正好是对方申请了patent的compound。对方的patent我
没看
到。文章看到了，就是culture里加compound,能杀死一些肿瘤细胞line。机理基本不清
。所以，我们的靶标和他们的没有任何关系。但业内人士说，我们拿到patent的可能性
不到50%。实际上，大学已经不愿意浪费钱，不要了。patent是我们自己找律师在搞。
正如你说的，如果compound真promising, 走non-profit是另外一条路。对我来讲，就
是有个饭碗。
