psin，phage等，ef1a, cag, pgk可以坚持一段时间，但是也存在被silence等现象.转
质粒总体比较困难，病毒是最高效的方法。在细胞传代的同时加lentivirus，
polybrene，然后spin半小时，效率会很高. rock inhibitor记得加，不然细胞会死光
光.病毒含血清的话几个小时后就可以换掉，如果要drug selection，过夜也没事，第
二天可以继续加rock inhibitor，浓度可以降到5um。

发帖数: 423 | 15 客观来讲，pfizer也是第一个吃螃蟹的人！目前来讲，CETP的inhibitor还没有上市的
药物，CETP还不是一个validated target.而且medicinal chemistry对该cetp的研究并
不深入，出现这样的问题也不足为奇了。毕竟不象当年lipitor,都是statin类药物相当
成熟的时候Warner-Lambert研究的类似物，pfizer直接收购的。
我想pfizer还是太担心lipitor的专利失效的问题了，可是心急吃不得热豆腐阿!结果栽
了！
现在的情况很可能是torcetrapib的化合物本身的问题，从我的对结构的分析看不象
CETP这个target出了什么问题。JTT-705目前作为CETP inhibitor还在研究。 |

我个人认为，如果将来要在高校搞科研，去读match的博后就挺好。
个人认为。。。
但是也不一定，就拿我老板为例子吧，做COPD-relevant protease inhibitors,做了好
多年发现好像走进了死胡同，每次申请grant,人家都觉得没有兴趣（我怀疑是idea太老
）而且因为一些客观原因，活性非常好的inhibitor拿去做动物实验也没有做成。
后来偶然的原因，老板和一些研究病毒的别的学校的教授联系上了，当我们把以前没有
活性的东西拿去screening,发现有些东西对其他病毒居然有抑制性。这让老版又看到了
拿grant的希望。。。
所以，做合成的，还是不要拘泥于某个方向，只要能做出有活性的东西，什么方向都不
错，尤其是方向好的方向。哈哈。
我随便说说的，MM斟酌着看。 |

，有效降低中风的风险。当然这也意味着它的主要副作用是增加出血风险。此外，吃
warfarin的病人的饮食要很讲究（avoid leafy greens)，有一些药物之间的反应，病
人还要经常monitor inr比较麻烦。
你说你父亲已经使用warfarin2年多，inr在target内，也没有什么不适（有没有仔细
monitor bleeding signs?），应该来说是manage的比较好的病人。
Dabigatran/Apixaban/Rivaroxaban是近年来出的新药。Dabigatran是direct thrombin
inhibitor, 后两个是factor 10a inhibitor。这些药的主要特性就是不需要频繁
Monitor inr，也降低了出血风险（跟剂量有关），而且还对预防中风更有效（或者non
-inferior)。价格自然也贵。
你父亲医生想给他换apixaban，有一个trial叫ARISTOTLE,评价这个药和warfarin在

disease. Most of the data are from phase II. I am not sure whether there is
any phase III data published. There is no data on head to head comparison
among these inhibitors.
There is no free lunch. Any drug has side effects. Not everyone experience
all the possible side effects. If your wife were treated with some inhibitor
, and experience severe side effect, the dose could be reduced or switched
to another one.

关于这个saw palmetto，它主要是用来治疗良性前列腺肥大的。
一个最新的报道提到它和finasteride (5alpha-Reducatase inhibitors)效果类似，但
副作用小。
另个报道说没效果：In two recent well-conducted trials of different saw
palmetto extracts, no effect on symptoms greater than placebo was seen, even
when the dose was pushed up to three times the standard dose of 320 mg
daily.
不过，临床医生对良性前列腺肥大的病人建议用此植物，因为基本无副作用。

因为治疗抑郁症的一线药物（SSRI，SNRI，Dopamine reuptake inhibitor）都是增加
脑内兴奋递质的，所以Depressed patients (who are actually bipolar but have
not yet had a manic episode), may switch into mania when given an 抗抑郁药，
比如 SSRI类药。而 Manic episode其中的一个显著表现就是spending sprees （疯狂
消费）, foolish business investments （胡乱投资）。和你很像。
建议你找专科精神科医生好好诊断和治疗。
另外，无论抑郁症还是Bipolar（抑郁症+躁狂），目前最有效的治疗方法是：
