家先用左手拿起筷子，lock，然后看看右手的筷子有没有free；偶数编号的哲学家先用
右手拿起筷子，lock，然后等左手的筷子，这样协调吃饭就不会有deadlock，当然这个
不是最优解法（懒得去想当时是怎么做的），因为有的哲学家吃了1000次，有的哲学家
只吃了1次，但是这个不仅避免了deadlock,也避免了starvation.
如果是两个threads 要处理一个shared resources， 可以用两把锁（L1, L2), 而且规
定必须是先获得L1, lock, 再获得L2, lock, （用round robin ?? - unsure) 然后处
理resources。如果一个thread 只看到L2 free, 规定它不能acquire L2.
这样判断对吗？或者请大牛说说更简单的方法？ |

ketone bodies rather than glucose during suckling stage, once they are weaned they would have to
convert to a glucose-dependent phenotype.
If somehow your KO mice have defect in glucose metabolism, they will die.
And they technically die of starvation.
Just a hypothesis. If that hits your case, it will be interesting. |

proliferation,就是MTT assay做出来的。我就去重复了，atcc新买的细胞，各种浓度
，时间，serum starvation, collagen coating，mtt, brdu,能试的都试了，什么效果
也看不到。
给那个组发信，没有任何回复。给合作小组写信，合作组说他们也什么都没有看到，而
且那个分子理论上应该被cancer research的cell line很快的降解掉，所以应该是什么
效果也看不到。

千个基因中哪些表达发生改变。简单点说，改变A是原因，我们来找谁是结果。
但是在我的case里，我是已知我要的结果或者说是phenotype，即某RNA processing不
正常时
的状态。我需要找的是所有会导致这个状态的基因。换句话说，也就是为我感兴趣的结
果找原因。
我今天和lab里了解array的人谈，他们给不出好的建议，因为他们都说现在array都是
用来做上述

一看好多细胞被洗掉了,但是奇怪的是正中间的不掉,贴壁一圈不掉,而是周边的细胞都
没了, western染色之后看起来像个'回'字. 具体样子在附件里. 我确定的一点是之后
的treatment 和western步骤并没有显著的让细胞减少, 所以貌似问题在用plain
medium 洗细胞和serum starvation步骤上. 是这样的吗, 怎么避免呢, 洗的时候很
gentle了, 怎么可以更gentle呢? |

food tracking.So, separtion of alcohol and food is not that straignt forward
in flies.
On the other hand, quantification of food intake has to be done after
starvation, which might complicate the feeding behavior after sex. |

phospho信号, 不知道有没有什么试剂是可以诱导明显的p38磷酸化增加,这样我可以用
来做positive control. 抗体用的是cell signaling的. 用cell signaling 的cell
extract跑western, 这个抗体可以很好的识别phospho-p38.

起过她以前的文章，但是那些文章都是一些计算分析为主的，所以我也比较不怎么当一
回事。昨天她谈了她们做的一些关于防止真菌进化出抗药性的实验，HSP90
抑制剂对于真菌没有直接杀伤力，但是综合和其他有直接杀伤力的药一起用的时候可以
非常好的防止真菌出现抗药性而复发。她最后的结论是HSP90的活性对于容忍机体的进
化有重要作用。高活性的HSP90会导致突变体能够成功存活下来。听完之后我忍不住跑
上去问了她几个问题，其中一个就是关于挨饿的问题，因为在starvation的时候据说那
些HSP活性会升高，那这样是不是会导致群体里面出现突变的几率升高？她毫不犹豫地
就说会，并说她们在酵母里面已经看到了类似现象了。然后我问了她有关HSP 在癌症
metastasis 里的因素的问题，她说她们正在做。
我其实还有另外一个问题，就是有关HSP90掩盖表型的问题，因为高活性HSP90虽然能够
导致突变体能够成功存活下来，但是貌似也会在表面上使得其表型趋于中性化? 如此说

一求教
1：如何定量。大家用软件还是手动数，软件的话有什么软件推荐？
2：还是定量问题：看到说autophagosome大小在0.5－1.5uM，照片上的很多小点是小于
这一值的，是不是只数大于0.5uM的？
3： 怎么解决heterogeneity的问题。看autophagy文章error bar都很小。怎么培养细
胞才能减少对照细胞的autophagy？ 同样的问题在starve之后也存在。
4：细胞在EBSS里starve6小时，也没看见GFP puncta明显增加，是不是有什么没有注意
到的问题？还是在这个荧光定量的实验中，大家都是默认加inhibitor的比如bafA1，即
使实验方法中没有明确说明？

第二，有些蛋白的mRNA是过量的，平时只有少量的会被翻译，其他的放着备用，在
stress response 或者细胞分裂的时候才会用上。所以你在平时来检测蛋白含量是看不
出来区别的。如果要确认这些问题，一个最简单的方法是用serum starvation --> add
back serum 的方式来激发细胞来使用大部分mRNA |

文章。
应该指出的是，常山即使在中药里，也是被视为一种虎狼之药的，因为其副作用太大，
属于在迫不得已的情况下才会用的药（疟疾在古代算是迫不得已的病情了）。但是它对
疟原虫的确是有效果的，这个也是不能否认的。葛洪收集的那个方子里，42个方子中有
14个还是13个是用这个常山的，这个可以说明那些方子并不是完全胡说八道的。

stock的情况下
承受track day的workload。所以要没有大的oil starvation overheat或者引擎
transmission可靠性问题。原厂suspension要比较好，之前看了996tt感觉很好，但去
论坛一查不砸个1w handling也就一般。
目前看了看e92 m3不错，11年后的貌似解决了rod bearing的问题。原厂稍微有点under
steer，但以后换轮胎size就能调整。不知大家有没有别的推荐？ |
