只是觉得这涉及很多的sequencing/genomics/bioinformatics,一方面很有趣；另外好
像找工作（不管是学校还是industry）是不是比较有前景？
所以想接受这方面（sequencing/bioinformatics)的训练
另外我在烂校，老板MD-PHD的AP；但老板的老板是超级大牛
按照你的说法，什么方向都是人满为患，大家都不用做了。
而且我读了一些paper，现在published的各种疾病的whole-genome structural
variants的研究，比较靠谱的结果还仅仅是基于array CGH的成果；而不是next-gen sequencing，因为一方面next-gen sequencing还是比较贵的，做不了大规模;另外基于sequencing data的各种鉴定软件的结果根本都自相矛盾，还需要软件进一步的开发当然更重要的是测序技术本身的发展（比如更大的read length）
当然了，我上面说的估计牛人们早开始谋划了，只不过还没publish出来
个人想法，说的不对请指正

牛实验室估计是几年有个CNS，剩余的实验室（大多数）其实就是发PNAS，structure，
JBC，JMB，biochem。所以说其实所有分支都差不多吧，没有谁好发文章或者谁不好发。
施一公，颜宁这次确实是手慢了。Anyway，总不能啥结构都被他俩抢走吧。。。

发帖数: 153 | 8 Rosetta第一步也是基于PDB database的homology modeling，然后再用force field做
计算。Rosetta近年在蛋白计算届几乎是最火的软件，发了大量文献，baker组资源强大
，当然宣传也做得很好。
不过确实需要些基础Linux知识安装很运行，以及一台还可以的机子（记得以前组里的
人试过，12 core 的机子跑3-5天，from sequence to refined structure)。网上资料
现在做得应该更完善了，记得还有Robetta web server 应该是可以帮你

发帖数: 1704 | 9 你说的对，Resetta后来也推出了基于Homology modelling的模块，不单单只靠ab
initio presiction的算法，但和传统意义上的homology modelling经典方法还是有所
区别的。
Baker的牛文确实层出不穷，他提出的这套方法原创性太强，基本上这个领域就看他一
个人在做，所谓“走自己的路，让别人无路可走”。
如果我没记错，近几次CASP的结果，rosetta基本都比不上I-Tasser（或大致持平？）
，等今年CASP11的结果出来再看。前面提到过，其实rosetta眼下的亮点和强项，在
structure refinement以及protein design上，这一块确实没有敌手，尤其是后者，让
Resatta大热。但眼前为数不多的几个成功范例能否再进一步推广，有多少普遍意义，
可能还不好说。
Robetta server基本就是个joke，你可以试一下普通权限在线提交一个>100aa的序列，
看看系统计算出来大约需要多久能给你结果...而Rosetta的帮助文档做的是出了名的烂
，这么久以来一直就没变过... |

是methodology；然后1000genome测了几千个正常人的SV
但我搜了半天都几乎没看到有什么具体的disease（貌似有几个cancer tumor的），说
某个病通过WGS寻找到新的SV的。（已知SV然后验证的不算）
或许是我搜索能力太差？

Trytophan fluorescence spectroscopy could be used to analyze whether protein
is compact or open. Ex at 288 nm and em around 320 nm suggests that protein
is compact (trytophan in hydrophobic environment). Em at longer wavelength
suggests an open structure. Also relative fluorescence intensity could tell
something? Anybody know why?
