why don't we just extract the hematopoietic stem cells from the patient,
introduce the delta32 mutation with homologous recombination (this is tricky
because you need to introduce dthis mut on both alleles, which I don't know
is achievable yet) and put the damn thing back to his own body? you don't
have to worry about the HLA compatibility at all.

接近量产车的。
B组时代虽然理论上也需要200辆来Homologation, 但是因为改装的潜力几乎是无限的，
造成拉力车跟量产车差别非常大，那两百辆街车基本都是怪兽。举例来说Lancia在B组
时代的037和DELTA S4，名义上是Monte Carlo和Delta的高性能版本，但是基本上跟大
量（远大于200辆）生产的Monte Carlo和Delta毫无关系。
quattro当时算是B组时代比较特殊的和量产车比较接近的，因为本身Quattro是当作高
性能跑车来设计的，境界跟其他的平民级别不一样。类似的有后来的Porsche 959和

后自己可能开展的研究课题或工作。我实在是不知道该怎末下手，我的背景是以前做过
神经元甘氨酸受体通道的电生理研究，后来做过GABAA受体还有protocadherin蛋白的研
究，但我对老年痴呆之类的一点概念都没有，现在上网查也只能了解各大概，也没找到
老年痴呆和我熟悉的GABAA受体或是protocadherin之间有关系的相关文献，有没有高人
指点一下该怎末入手设计这个面试课题啊，万分感谢。
补充一下，先前通email他们好像对我的电生理研究背景比较感兴趣，另外我熟悉的实
验技术包括细胞培养，免疫荧光，western blot，基因克隆等。还有我查了一下，他们
研究所有人做β-Site APP Cleaving Protein 1 (BACE1)和CutA Divalent Cation
Tolerance Homolog (E. coli) (CUTA)的研究，还有人做AD小鼠模型。

转行篇：
先介绍一下我的背景吧，本科是国内重点高校数学专业基础方向，当然这个方向的意思
是选修几门基础数学方向的课。博士是美国100左右（应该是开外的）的烂校数学专业
拓扑方向。之前 的cs背景是本科学了一学期的c语言（本科混混的，所以基本等于没学
）。2010年的时候，自学了一点iphone programming，但是也不深入，就是一点皮毛吧。
大概是2011年的4月份，我的phd第四年即将结束。我走进老板办公室，和他讨论毕业的
事情。那时候我的博士论文已经做得差不多了。已经发表和即将发表的论文一共三篇。
作为一个烂校基础数学的学生，这个差不多可以毕业了（不能和cs的牛人动不动就10几
篇的比，我们基础数学专业要求不高，不用很多论文）。在2010年4月的时候，我们讨
论同一个话题的时候，我老板数着手指头说，明年第五年，应该可以毕业了。但是，计

看了Zhang Feng的Nature Protocol关于CRISPR的文章，Figure 6里面的ssDNA
template明显的效率比plasmid作template高。文章并没有明确说明哪个template好。
想了一下，ssDNA分子量比plasmid小多了，同样的ug template, ssDNA的分子数肯定多
多了，也许这是一个解释。
我自己在设计HDR template，想两边各一个Kb的homologous arm,中间有60-70bp 的
mutation insertion。这个两kb的DNA准备克隆到一个plasmid里面，sequencing确定
mutation后，就可以用作HDR template了。
