面我的人是team的director，把resume上的经历过了一遍，问的很详细，连在学校做TA
的都问了。刚打电话让我下周再面一次，下回是两个人，上回的director+另一个，问
了问他们是technical interview还是personality fit，他们说director会偏向问
personality的东西，另一个人偏向technical。
之前从来没面过technical的，想问问有没有interviewer或者被面过technical
question的大神们指点一下，可能会出什么样的问题？

指标还是很重要的。
烧，最好是都做，即sample prep也做technical replicates，然后到sequencing时，
对上面的两者分别再做technical replicates。不过据我的经验，technical
replicate现在已经意义不太大了，有这钱还不如多做几个biological replicate。 |

发帖数: 1 | 5 最近做luciferase assay，就是把含有疾病相关的snp的promoter跟luciferase连接在
一起，看snp对promoter的影响。假设snp的ref allele是A, 疾病相关的allele是B
我首先在hek293里做，用lipofectamine3000转染。铺板子（12-well）铺了10个孔，5
个用于allele-a, 5个用于allele－b。先把lipo和dna混合好，然后把mix分给每一个孔。
以上是第一次实验，过两天我会一模一样的重复上述实验。
我想请问这里的5个孔中的每一个孔算是biological replicate还是technical
replicate呢？
而这两次实验算是biological replicate还是technical replicate呢？
之所以问这个问题是因为后来面对转染neuron的实验。neuron比较难转染所以用的是
nucleofection，一次性用cuvette转染100万个细胞，然后分到12-well的3个孔中。
同样的实验过两天我也会一模一样的再重复一次。

我们组主要负责Dropship核心业务的处理。Dropship业务最近发展很快，业务量，业务

环节不同而已。文章中更侧重的是sequencing本身这个部分，你所说的则是sample
prep过程以及sequencing，但如果出问题不容易确定原因在哪个环节。所以如果有钱烧，最好是都做，即sample prep也做technical replicates，然后到sequencing时，对上面的两者分别再做technical replicates。不过据我的经验，technical replicate现在已经意义不太大了，有这钱还不如多做几个biological replicate。 |

是希望知道technical replicates 的variation。
比如有人想做基因表达普，找差异表达的基因。一般现在第一个问题就是用microarray
还是 RNA-seq。如果目的是找差异表达基因，那么variation between technical
replicates 直接影响到 statistical power。如果同样两份细胞，通过 RNA-seq 流程
做出来（包括建库）的variation 比通过 microarray 流程做出来的variation 大，那
就是用microarray 做更好了。
那种之比较不同 lane 之间的 variation 对生物学家来说，没有什么现实指导意义。
另外，现在不少人认为 RNA-seq 成本已经将下来，和microarray 差不多了。可是这个
成本的下降是通过barcode multiplex 来实现的的。所以要比较 RNA-seq 和
microarray，最好能在同成本的条件下比。 |

需要你纠结的时候，就说明这种提法不够准确了。
其实考虑这些问题非常的直接，你的统计学结论的scope是什么。你一次
铺5个，最后能做出的结论就是“9月12日我的实验结果说明a和b不一样“；
你第二天再搞一次，结论就是”9月12日和13日的实验结果说明a和b不一样“。
其实你想要的结论是“a和b不一样”，你所有的实验都没法推出这个结果，
你从前面两个实验的结果来推，这叫做extrapolation. 要注意，extrapolation
都是不靠谱的，当然你做两次试验比只做一次相对靠谱一点。至于啥biological/
technical都没啥区别，你第一次的实验想对mix好了测5次也算是biological，
你第二次试验相对于重新买一批hek293重做也算technical, 程度不同而已
